| dc.description.abstract | In this study, the morphological and microscopical properties of the specimens from the species, Lactarius deliciosus, Lactarius sanguifluus, Lactarius semisanguifluus,Lactarius deterrimus, Lactarius salmonicolor belonging to Lactarius genus, Russulaceae family and Russulales order were examined and also protein profiles of these specimens were analysed by PAGE. In this investigation, morphological similarities and differences of these species were determined both by botanical excursions and by laboratory studies. Microscopical examination of cuticle and spores were done.According to structure of cuticle, it was observed that ; Lactarius deliciosus, Lactarius deterrimus and Lactarius salmonicolor are belong to monomitic hyphal system structure Lactarius sanguifluus and Lactarius semisanguifluus are belong to dimitic hyphal system structure. Both SDS - PAGE and N - PAGE analysis were demonstarted that these species have different protein band profiles. The five species which the morphological properties are very similar, were identified by the structure of their cuticle and also shown by PAGE analysis that they are completely different species from the view of molecular typing. | dc.description.ozet | Bu çalışmada, Russulales ordosu Russulaceae familyası Lactarius cinsi mensubu olan; Lactarius deliciosus, Lactarius sanguifluus, Lactarius semisanguifluus,Lactarius deterrimus, Lactarius salmonicolor türlerine ait örneklerin makroskobik ve mikroskobik özellikleri incelendi, aynı örneklerin protein profillerinin PAGE ile analizleri yapıldı. Araştırmamızda bu türlerin morfolojik olarak benzerlikleri ve farklılıkları hem arazi çalışmalarıyla hemde laboratuvar çalışmalarıyla belirlendi.Kutikula ve sporlarının mikroskobik incelemeleri yapıldı.Kutikula yapılarına göre Lactarius deliciosus, Lactarius deterrimus ve Lactarius salmonicolor' un monomitik hifsel sistem yapısına, Lactarius sanguifluus ve Lactarius semisanguifluus' un ise dimitik hifsel sistem yapısına sahip olduğu gözlendi. Gerek SDS - PAGE gerekse N - PAGE çalışmalarında incelenen bu türlerin farklı protein bant profillerine sahip olduğu gösterildi. Morfolojik özellikleri birbirlerine çok benzerlik gösteren bu beş türün kutikula yapılarına göre birbirlerinden ayrımları sağlandığı gibi moleküler tiplendirim açısındanda tamamen farklı türler oldukları PAGE analizleri ile ortaya konuldu.ANAHTAR KELİMELER : Lactarius, mantarlar, sınıflandırma, morfoloji, anatomi, protein, elektroforez | tr_TR |
