Abstract

The genus Eryngium L. is a cosmopolitan and the most species-rich genus of Apiaceae. It is used by local people as antiinflammatory, antioedema and against pain, scorpion and snake bites. It is also used in Anatolia in similar ways. In our study, the ethanolic and water extracts prepared from the aerial and underground parts of eight Eryngium species (Eryngium campestre L., Eryngium creticum Lam., Eryngium davisii Kit Tan&Yildiz, Eryngium falcatum Delar., Eryngium isauricum Contandr.&Quezel, Eryngium kotschyi Boiss., Eryngium maritimum L. and Eryngium trisectum A.Wörz & H.Duman) were examined antinociceptive activity using p-benzoquinone-induced writhing test and for their anti-inflammatory activity by using carrageenan-induced hind paw oedema and TPA-induced ear oedema tests which were employed on mice. Eryngium kotschyi was found to exhibit higher activity in both tests. The methanolic extract prepared from this plant?s underground parts, was partitioned with hexane, dichloromethane and n-butanol (water saturated) respectively; after evaporating the solvents, the crude extracts were applied to acetic acid induced writhing test and hot plate test in order to evaluate their antinociceptive activity. The n-butanol extract was found to possess significant activity, after which isolation studies continued with this fraction. Main fractionation was carried out using normal phase silica gel column chromatography; obtained subfractions were tested again to establish the active fractions. The fractions which showed high activity were subjected to multiple chromatographic steps (normal phase silica gel MPLC and RP-18 MPLC) in order to isolate four triterpenoid saponins. Their structures were elucidated mainly by 1D (1H-NMR and 13C-NMR) and 2D (1H-1H COSY, TOCSY, NOESY, HSQC, HMBC) NMR spectroscopy and FAB-Mass spectroscopy. Their structures were determined as 3-O-?-L-rhamnopyranosyl-(1?4)-ß-D-glucurunopyranosyl-22-O-angeloyl-A1-barrigenol; 3-O-?-L-rhamnopyranosyl-(1?4)-ß-D-glucurunopyranosyl-22-O-ß,ß-dimethylacryloyl-A1-barrigenol; 3-O-ß-D-glucopyranosyl-(1?2)-[ß-D-glucopyranosyl-(1?6)]-ß-D-glucopyranosyl-21-O-acetyl-22-O-acetyl-28-O-acetyl-3ß, 21ß, 22?, 28-tetrahydroxyolean-12-ene-16-oxo and 3-O-ß-D-galactopyranosyl-(1?2)-[?-L-arabinopyranosyl-(1?3)]-ß-D-glucurunopyranosyl-22-O-angeloyl-A1-barrigenol which they were all found to be structurally new. Since amount of the compounds was not sufficent, only 3-O-ß-D-galactopyranosyl-(1?2)-[?-L-arabinopyranosyl-(1?3)]-ß-D-glucurunopyranosyl-22-O-angeloyl-A1-barrigenol was examined for its antinociceptive activity by hot plate test. As a result, it was found to possess a moderate antinociceptive activity.
