Abstract

In this study, active compounds of the sponge sample of Agelas oroides from the Mediterranean sea in our country have been isolated. DPPH-radical scavenging activity and acetylcholinesterase enzym inhibitory effects of the sponge extract and active compounds have been investigated.Sponges were collected from the coast of Kemer from 25 meter depth in August 2007 and the extract was prepared with methanol. Starting with the methanol extract, ethyl acetate and hexane fractions were obtained; oroidin and 4,5-dibromopyrol-2-carboxylic acid from ethyl acetate phase and 25-hydroxy-24-methylcholesterol from hexane fraction were isolated as the major compounds. TLC, column chromatography, HPLC and LC-MS methods were employed during isolation.Activity studies were performed after isolation. In the first phase of activity studies, DPPH-radical scavenging activity of the methanol extract and isolated compounds have been determined. DPPH-radical scavenging activity has been assayed as 29.76 ± 4.66% for dilution of 2 mg/ml of methanol extract of the sponge, 93.91±0.46% for dilution of 2 mg/ml of oroidin, 16.58±0.89% for dilution of 2 mg/ml of 4,5-dibromopyrol-2-carboxylic acid and 6.44±0.97% for dilution of 2 mg/ml of 25-hydroxy-24-methylcholesterol.In the second step of the study, acetylcholinesterase enzym inhibiting activity of the methanol extract and isolated compounds have been investigated. It was found that 2 mg/ml dilution of the methanol extract of the sponge showed 47.02±4.40% enzym inhibiting activity whereas 2 mg/ml dilution of oroidin showed 55.21±2.94% and 2 mg/ml diluiton of 25-hydroxy-24-methylcholesterol exerted 7.85±2.42%. 4,5-dibromopyrol-2-carboxylic acid did not show any activity.
