Abstract

The fluorescent appearance and titreş of antibodies formed against cytoplasmic structures during early pregnancy in dogs In this study, it was aimed to reveal the differences between pregnant and non-pregnant bitches with regard to the titreş of antibodies (antinuclear and cytoplasmic) and fluorescent appearances (netlike, filamentary and perforated). It was also aimed to determine whether there is a difference between pregnant and non-pregnant bitches as regards progesterone and oestrogen values. The animal samples needed for the study were obtained from the Central Headquarter of Dog Breeding and Raising of 4th Main Headquarter of Jet Base (KÜVEM) and from various races of healthy dogs in the Private Farm of Academia For Dog Breeding (German shepherd dog n=7, Golden Retriever n=1, Siberian wolf n=1, Rotvider n=2, Doberman=1, Labrador n=1, Setter n=1). Bitches were dividen into two groups. Group I ( n=7) consisted of normally pregnant female bitches ( 5-35 days after mating). Group II ( n=7) consisted of the bitches which could not become pregnant bitches because they were not allowed to mate after displaying oestrus (5.-35 days, beginning of metestrus). In serums, with AAb Indirect Immune fluorescent (IIF), Canine Ana Hep-2 Screening Kit (Inova Diagnostic Inc.) were analysed. Human epithelial cell line was used as Hep-2 substrate and Anti-canine IgG was applied as conjugate. They were marked with fluorescent isotiyosiyanat (FITC) (in Nordic Immunology, Netherlands, 1:1280 BS). The display of serum samples were carried out in accordance with the directives of the producer company. According to the valid fluorescent type, the fluorescent samples of the cytoplasm (Pollak et al, 2001b). Type-1 is like netlike, type-243 filamentary and type-3 perforated cytoplasm. Pregnant and non-pregnant were differentiated according to these formations. While there was no statistically significant difference detected between pregnant and non-pregnant bitches with regard to the titreş of antinuclear antibodies (ANA) on 5/6 and 24/25th days, remarkable differences were observed on 10/11 and 35/37th days with p<0,05,and on 15/1 6th days p<0,01. It was determined that the differences was not significant as regards fluorescent differences. No statistically difference has been detected in pregnant and non pregnant bitches with regard to either progesterone or oestrogen values. An important finding in hormonal values is that while progesterone values drop on 35/37th in days in pregnant bitches remarkably (p<0,03), oestrogen values increased in pregnant bitches significantly (p<0,01). In all the pregnant bitches observed in this study different cytoplasmic fluorescent was detected, which was formed due to AAb despite possible structural proteins. In all samples, negative chromosomes were striking. The findings obtained indicated that pregnancy differs at different stages regarding ANA titreş and that immunological tests are important in determining the physiological difference of pregnancy.
