Abstract
Recently, various types of stem cells have been widely used as vehicles to distribute the therapeutic genes to metastatic deposits in cancer gene therapy. Mesenchymal stem cells (MSCs) have been widely used for this purpose. The improved migration of mesenchymal stem cells (MSCs) to the tumor tissues has made them an attracticve vehicle for gene therapy. Therapeutic genes can be loaded by using plasmids and viral vectors. Adenoviruses are popular gene delivery vehicles in cancer gene therapy because of its transient episomal expression in mammalian cells and no risk of mutagenesis. However, they can not easily transduce the MSCs. Very recently, due to lack of toxicity to the mammalian cells, the baculo viral vectors have been studied extensively in experimental cancer models. Baculoviral vectors seem to have advantage of efficient transduction of MSCs. In this study, we aimed to construct recombinant baculoviral vectors carrying cytosine deaminase (CD) or/and GM-CSF transcription units and to load these vectors to the bone marrow mesenchymal stem cells. Finally, we tested those vectors in a mouse model of colon carcinoma.
