Abstract

Ochratoxin A (OTA), a mycotoxin produced by several fungi of Aspergillus and Penicillium species in the food chain, has nephrotoxic, hepatotoxic, carcinogenic, teratogenic and immunotoxic effects. It is also claimed that ochratoxin A mostly causes Balkan Endemic Nephropathy (BEN) seen in Southeastern European countries. OTA is classified as a possible carcinogen to human (Group 2B) by the International Association of Research on Cancer (IARC).In this study in order to define the OTA occurrence and level in human urine, NaHCO3 dilution, IAC clean-up and HPLC fluorescent detection and definition methods were used. As a result of the validation study for the analysis method that includes the parameters like repeatability, trueness, selectivity, accuracy for the limit of quantification and limit of detection for linearity the correlation factor is r=0.9922, for precision, worked on 5 level spiked samples as 0.02, 0.05, 0.1 and 1 ppb, the RSD (%) was found as 3.83, 7.32, 6.91, 3.29 and 8.86 respectively. On the same spiked samples for the trueness study, the recovery percentage rates were found as 100.1, 110.5, 85.7, 92.5 and 88.6 respectively. (LOD and LOQ) of the method were measured as 0.006 and 0.018 ng/mL respectively. According to these results the parameters related to the performance test for EC regulation of 401/2006 were found suitable.For the analysis, first morning urine samples were collected from the voluntaries and the results were adjusted with creatinine levels. The urine samples were collected from 4 different cities as Ankara, İstanbul, Diyarbakır and İzmir. From the total collected samples of 233 larger amount of 83% was contaminated with OTA. Among the measured OTA levels, positive sample rate, average OTA amount and the highest contamination was found in Ankara. (Positive sample rate; 90.1%, average OTA concentration; 14.34 ng/g creatinine and highest OTA value;75.60 ng/g creatinine). The OTA contents of the urine samples collected from Ankara were higher than the OTA contents of the urine samples of Diyarbakır, Istanbul and İzmir (p<0.003).In order to define the exposure profile to OTA in human a questionnaire was conducted among the voluntaries as well. But after the evaluation of the answers related to the gender, age, dietary habits, coffee consumption, smoking and alcohol habits, no correlation was found with the OTA exposure.
