dc.description.abstract | In this study, 388 cloacal swab samples were collected from 109 poultry flocks, 106 bovine faecal samples and 107 faecal samples were collected from 27 sheep herds. Total of 601 faecal samples of different animals were examined for the existence of Salmonella spp. in this study. For the isolation of Salmonella, nonselective preenrichment by TPS was followed by selective enrichment by TTB and plating on selective agar XLT4. Suspect colonies were confirmed biochemically and serologically. Salmonella was isolated in 11(10.09 %) of the 109 broiler flocks tested. Of the 12 (3.09 %) Salmonella strain identified from 388 (10 swabs/sample) poultry cloacal swap samples, 9 (75 %) were detected as serogroup B, and 3(25 %) were detected as serogroup D. From the total of 106 bovine faecal samples, 1 (0.94 %) was isolated as Salmonella serogroup B. Salmonella was not isolated from 107 sheep feacal samples collected from 27 sheep herds. Isolates taken from tetrathionate cultures and determined as Salmonella spp. after biochemical and serological tests were also tested by PCR with primers specifically amplifying 606 bp portion of iroB gene. In all of the Salmonella positive cultures Salmonella spp. specific PCR products were detected. In this study, prevalence of Salmonella strains in animal feacal and cloacal swab samples was determined by using conventional technique. As a result, PCR technique is found to be useful for routine examinations of Salmonella in animal faecal samples since all Salmonella spp. isolated samples were confirmed by PCR. | tr_TR |
dc.description.ozet | Bu çalmada, 109 tavuk sürüsüne ait 388 adet kloakal svap örnei, 27 koyun sürüsünden 107 adet ve srlardan ise 106 adet dk olmak üzere toplam 601 adet örnek Salmonella varl yönünden incelenmitir. %zolasyon çalmalarnda, ön zenginletirme için Tamponlanm Peptonlu Su (TPS), selektif zenginletirme için Tetrathionat broth (TTB) ve izolasyon için selektif besiyeri olan Xylose Lysine Tergitol 4 (XLT4) agar kullanlmtr. Salmonella üpheli bulunan kolonilerin identifikasyonu, biyokimyasal ve serolojik yöntemlerle gerçekletirilmitir. Çalmada, incelenen 109 broyler sürünün 11 (%10,09)'inden Salmonella izolasyonu yaplmtr. 3880 klokal svaptan hazrlanan 388 (10 kloakal svap 1 örnek) adet örnekten identifiye edilen 12 (%3,09) adet Salmonella izolatnn 9 (%75)'u B serogrubu, 3 (%25)'ü de D serogrubu olarak tespit edilmitir. Srlardan toplanan 106 adet dk örneinden yaplan ekimler sonucu 1 (%0,94) adet B serogrubuna ait Salmonella izolasyonu yaplmtr. 27 koyun sürüsüne ait 107 adet koyun dk örneinden yaplan incelemede Salmonella tespit edilememitir. Biyokimyasal ve serolojik testler sonucunda Salmonella izole edilen örneklerin tetratiyonat buyyon kültürleri ile IroB gen spesifik primerlerle PCR gerçekletirilmi ve sonuçta kültür ile pozitif elde edilen tüm örneklerde Salmonella cinsine özgün 606 bp büyüklüünde PCR ürünleri saptanmtr. Sonuç olarak bu çalmada, hayvan dklarndan ve kloakal svaplardan Salmonella etkenlerinin kültür ile prevalans saptanmtr. Ayrca, Salmonella izole edilen tüm örneklerin PCR yöntemi ile de benzer sonuç vermesi, PCR'nun Salmonella etkenlerinin dklarda aratrlmasnda rutin tehis amacyla uygulanabilirliini ortaya koymutur. | tr_TR |
