    Köpek ve kedilerde deri ve eklenti tümörlerinin patomorfolojisi

    Bu çalışmada; 2009-2012 yılları arasında, Ankara Üniversitesi Veteriner Fakültesi'ne ve Ankara'da bulunan veteriner kliniklerine deri tümörü şüphesiyle getirilen 43 köpek ve 4 kediye ait toplam 47 doku örneği sitopatolojik, histopatolojik ve immunohistokimyasal olarak incelendi. Sitopatolojik örnekler sedasyon altında İİAB yoluyla alındı ve preparatlar MGG ile boyandı. Histopatolojik inceleme amacıyla cerrahi operasyonla alınan doku örnekleri formaldehitte tespit edilerek rutin doku takibine alındıktan sonra parafine gömüldü. Parafin bloklardan 5 mikron kalınlığında kesitler alınarak hematoksilen-eosin (HE) ile boyandı. Aynı bloklardan hazırlanan kesitler sitokeratin ile immunohistokimyasal olarak işaretlendi. Tümörlerin alındığı köpeklerden 29'u, kedilerden 2`si erkek olup, 14 köpek ve 2 kedi ise dişiydi. Tümör görülme yaşının köpeklerde 8-15, kedilerde 12-13 yaşları arasında arttığı gözlendi. Köpekler 14 terrier, 14 melez ve 15'i diğer ırklardandı. Kedilerin hepsi ise tekirdi. Tümörlerden 10'u benign, 37'si ise maligndi. Tümörler lokalizasyon olarak daha çok kafa, perianal bölge ve ön-arka bacaklarda yerleşim göstermekteydi. Sitopatolojik ve histopatolojik olarak; 14 yassı hücreli kanser, 9 hepatoid bez tümörleri, 8 bazal hücreli kanser, 7 ter bezi tümörü, 5 sebaseöz bez tümörü, 2 papillom, 1 malign trikoepitelyom ve 1 malign pilomatrikom tanısı konuldu. Çalışmada sitopatolojik olarak %84.4 doğru tanıya gidilmiştir. Malign tümörlerde hücrelerde pleomorfizm, dev hücreleri, çekirdek anomalileri, çok çekirdeklilik ve atipik mitoz gibi malignite kriterlerine rastlandı. Histopatolojik olarak tümörler Dünya Sağlık Teşkilatı (WHO) Evcil Hayvanlarda Deri Tümörlerinin Histolojik Sınıflandırılmasına göre tanımlandı. İmmunohistokimyasal olarak bütün tümörler sitokeratin antijeni için pozitifdi.AbstractIn this study, a total of 47 (43 dogs and 4 cats) skin tumors were diagnosed by cytopathological, histopathological and immunohistochemical. Tissue samples submitted from Faculty of Veterinary Medicine Ankara University and private clinics of veterinary in Ankara during a two years period from 2009 to 2012. Cytologic specimens were prepared by Fine Needle Aspiration Biopsy (FNAB) and stained with May-Grunwald Giemsa (MGG). For histopathologic examinations tissue samples were obtained by surgery. Samples were fixed in formalin and embedded in paraffin by routine procedures. Paraffin sections of 5 micron thickness were stained with hematoxylin-eosin (HxE), and immunperoxidase method was performed by cytokeratin. Tumors were obtained from 29 dogs and 2 cats male, 14 dogs and 2 cats female. The age with highest incidence in dogs were between 8?15 in cats were 12?13. The more frequent affected breeds of dogs were 14 terrier, 14 mongrel and 15 another different breeds. All of cats were mixed-breed. The total number of skin tumors were 47; from those 10 benign and 37 malignant. Localizations of tumors were in the followed regions, head, perianal regions and legs. There were cytopathologycaly and histopathologycaly diagnosed were including 14 squoamous cell carcinoma, 9 hepatoid gland tumors, 8 basal cell carcinoma, 7 sweat gland tumors, 5 sebaseöz gland tumors, 2 papilloma, 1 malignant trichoepithelioma and 1 malignant pilomatricoma. In the study the cytopathologic diagnosis was in agreement %84.4. In the malignant tumors polymorphism, giant cells, nucleus with anomalia, binucleated cells and mitosis were seen. Histopathologically tumors were classified according to WHO normative and the classification scheme proposed by Goldschmitd et all. (1998). İmmunohistochemically all the tumors were positive for cytokeratin.

    Proliferative and apoptotic evaluations of renal preventive effects of coenzyme Q10 in radioiodine-131 induced renal damage

    The aim of this study was to investigated anti-proliferative and anti-apoptotic effects of coenzyme Q10 (CoQ10) in the prevention of radioiodine-131 (RAI) (I131) induced kidney damage. A total of 24 Wistar albino rats were separated into equal three groups (n = 8/group): Group 1 (control): untreated group; Group 2 (RAI): 3 mCi/kg RAI oral route; Group 3 (RAI+CoQ10): 3 mCi/kg RAI oral route and intraperitoneally 30 mg/kg/day CoQ10. CoQ10 treatment was started two days before RAI administration and was continued five days once daily after RAI. Pathomorphological parameters of kidneys were measured using hematoxylin–eosin and Masson’s trichrome staining. Immunohistochemically; proliferating cell nuclear antigen (PCNA), caspase 8, caspase 9 and terminal deoxynucleotidyl transferase–mediated deoxyuridine triphosphate nick end labeling (TUNEL) were used to determine proliferation and apoptosis. With the exception of the control group, varying degrees of inflammation, degeneration, necrosis, and interstitial/perivascular fibrosis were detected in the kidneys of all rats. This histopathological damage was found to be significantly less in CoQ10 group versus RAI group (P<0.05). The all immunohistochemical examinations demonstrated that administration of CoQ10 had reduced proliferation and apoptosis (P<0.05). The results of kidney histopathology and immunohistochemistry demonstrated that administration of CoQ10 had reduced inflammation, proliferation, and apoptosis. These findings show CoQ10 can play an important role in the radioprotection of kidneys against RAI-induced damage
