Item Akciğer kanser hücresi ile insan alveolar epitel hücresinin etkileşiminin incelenmesi(Fen Bilimleri Enstitüsü, 2015) Ermiş, Ezgi Kaya; Cebesoy, SunaAkciğer kanser hücreleri, etrafındaki hücreleri etkileyen sinyaller oluşturarak bir Tümör Mikro Çevresi (TMÇ) oluşturmaktadır. Bu çalışmada; TMÇ etkileşimine dayanarak tümörlü hücrelerin, tümöre yakın dokuda büyüyen sağlıklı alveol epitel hücrelerin fonksiyonlarını nasıl etkiledikleri araştırılmıştır. Primer insan alveolar epitel hücreler (AT2), akciğer adenokarsinoma hücreleri (A549 ve H1975) ile ko-kültürü yapılmıştır veya adenokarsinoma hücrelerinden elde edilen koşullandırılmış besiyerine maruz bırakılmıştır. Alveolar bariyer gerginliğinin göstergesi olarak Transepitel Elektriksel Direnç (TEER) ölçülmüştür. AT2 hücre proliferasyonu kristal viyole ile ölçülmüştür. Ussing düzeneğinde Epitel Na kanallarının (ENaK) ve Na+/K+-ATPaz ölçümü ve bunların alt ünitelerinin mRNA ekspresyonlarının belirlenmesi ile Na+-iyon transportu değerlendirilmiştir. Glikoz ve laktat düzeyi metabolik faaliyeti belirlemek için değerlendirilimiştir. O2 bağımlı reaksiyonların rolünü değerlendirmek için normal ve gaz geçirgen plateler kullanılmıştır. Çalışmamızın bulgularına göre; koşullandırılmış besiyeri ENaK ile Na+/K+-ATPaz aktivitesini stimüle etmektedir ve TEER’i arttırmaktadır. Adenokarsinoma hücreleri AT2 hücrelerinin proliferasyonunu etkilememektedir Adenokarsinom hücreleri ile AT2 ko-kültürü TEER’i arttırmaktadır ve 24 saatlik stimülasyondan sonra ENaK ve Na+/K+-ATPaz aktivitesi azalmaktadır. Bu azalma O2 geçirgen plakalar sayesinde önlenmiştir. Adenokarsinoma hücre ile AT2 ko-kültürü, plate türünden bağımsız olarak her iki plate tipinde de glikozu azaltmış ve laktatı arttırmıştır. Adenokarsinoma hücreleri ile AT2 ko-kültürü 24. saatte β-ENaK alt biriminin mRNA ekspresyonunu arttırmıştır. Uzun süreli maruziyet, oksijenasyonun azalmasına neden olmuştur. Bu sonuçlar, adenokarsinoma hücrelerinin çözünür faktörler aracılığı ile sağlıklı AT2 hücresini stimüle ettiğini göstermektedir. Ayrıca, adenokarsinoma hücrelerine uzun süreli maruziyet, AT2 hücresinin fonksiyonunu hipoksi aracılığı ile azalttığı tespit edilmiştir.AbstractLung cancer cells generate a microenvironment by releasing signals that affect surrounding cells. Based on this interaction, it was investigated that cancer cells affect the function of healthy alveolar epithelium where growing in close vicinity of tumor tissue in this study. Primary human alveolar epithelial cells (AT2) were in co-culture with adenocarcinoma cells (A549 and H1975) or with conditioned media from adenocarcinoma cells. The transepithelial electrical resistance (TEER) was measured as indicator of barrier tightness. Proliferation of AT2 was measured with crystal violet. Na+-ion transport was evaluated by measuring of epithelial Na channels (ENaC) and Na+/K+-ATPase in Ussing chambers and by determining mRNA expression of their subunits. Levels of glucose and lactate was evaluated for assessing metabolic activity. Normal and gas-permeable culture plates were used to evaluate the role of O2 dependent reactions. According to our results, conditioned media increased TEER and stimulated ENaC and Na+/K+-ATPase activity. Proliferation of AT2 cells was not affected. Co-culture with adenocarcinoma cells increased TEER and after 24 hours stimulation decreased ENaC and Na+/K+-ATPase activity. This decrease was prevented by culture on gas permeable plates. Co-culturing AT2 with adenocarcinoma cells decreased glucose and increased lactate in the culture medium from both plate types independent of the type of culture plate used. Co-culture with adenocarcinoma cells increased the mRNA expression of β-ENaC subunit at 24. hours. Longer exposure caused a decrease of oxygenation. These results indicate that adenocarcinoma cells stimulate healthy alveolar epithelium mediated by soluble factors. Also, it was detected that prolonged exposure to adenocarcinoma cells decrease AT2 function by hypoxia.Item Bazı yarasa türlerinin uçma kaslarında morfolojik ve histokimyasal çalışmalar(Fen Bilimleri Enstitüsü, 1998) Cebesoy, Suna; Ayvalı, CevatMyotis myotis ve Miniopterus schreibersi'ye ait iki esas ve iki yardımcı uçma kası morfolojik ve histokimyasal olarak çalışılmıştır. M.myotis ve M. schreibersi ' nin pektoral kasında histokimyasal olarak iki tip hızlı kasılan fibril tanımlanmıştır. Glisin-kalsiyum-formaldehit preinkübasyonundan sonra miyozin- ATPaz ile boyanarak bunlar tip Ha ve tip Hb olarak sınıflandırılmıştır. Esas uçma kası olan serratus ventralis ve yardımcı uçma kasları triceps brachii ve biceps brachii tip I, tip ila ve tip lib fibrilleri içermektedir. Her iki yarasa türünde pektoral kas dışındaki uçma kaslarındaki bütün tip Ifibriller NADH-TR ile koyu boyandıklarından dolayı yüksek oksidatiftirler. Tip II a fibrilleri NADH-TR veSDH ile nispeten daha zayıf boyanmışlardır, buda orta derecede oksidasyon kapasitesini göstermektedir. M.myotis ve M. schreibersi esas ve yardımcı uçma kaslarındaki tip lib fibrilleri, NADH-TR ye zayıf reaksiyon verdiklerinden dolayı düşük oksidatif olarak kabul edilmişlerdir. M.myotis ve M. schreibersi'nin uçma kasları morfolojik olarak karşılaştırılacak olursa; M. schreibersi küçük bir yarasa olmasına rağmen fibrillerinin çapı M.myotis' den daha büyüktür. M. schreibersi 'nin yardımcı ve esas uçma kaslarının fibril çapları M.myotis 'de bulunan fibrillerin çaplarından daha büyüktür. M. schreibersi'nin fibril dağılımı, pektoral kasda tipli a fibrilleri % 87, tipli b fibrilleri ise %13, M.myotis' de ise bu dağılımın tipli afibrillerde % 76, tipli bfibrillerde ise % 24 oranında olduğu tesbit edilmiştir. Serratus ventralis'de, triceps brachii ve biceps brachii de hızlı fibrillerin yüzdeleri M. schreibersi' de, M.myotis'e göre daha fazla bulunmaktadır. Buna karşılık M.myotis' in seratus ventralis, triceps brachii ve biceps brachii nin tip I fibrilleri yüzde olarak M. schreibersi'nin aynı kaslarındaki tip I fibrillerinden daha fazladır.AbstractTwo primary and two accessory flight muscles of Myotis myotis and Miniopterus schreibersi were studied using morphological and histochemical analysis. Two fast-twitch fiber types are histochemically identified in pectoralis muscles of M.myotis and M. schreibersi. These were classified as type II a and type II b according to gly cine-calcium-formalin preincubation staining protocol for myosin ATPase. The primary flight muscles, serratus vent alis and the accessory flight muscles,triceps brachii and biceps brachii included type I, type II a and type II b fibers. All type I fibers in two bats flight muscles (except pectoralis) were highly oxidative, as stained darkly for NADH-TR. All type II (i fibersexhibited relatively intense staining properties for NADH-TR and SDH, suggesting anintermediate oxidative capacity. In M.myotis and M. schreibersi primary and accessory flight muscles, type II b fibers were low oxidative, as indicated by light reaction for NADH-TR. If flight muscles of M.myotis and M.schreibersi are compared depending on this study; Although Mschreibersi is a small bat, the diameters of it's fibers are greater than that of M.myotis. Fiber diameters of primary and accessory flight muscles of M.schreibersi are greater then M.myotis' s fiber diameters. Fiber ratios of M.schreibersi for the pectoral muscle are 87 % for fiber type II a and 13 % for fiber type II b. Fiber ratios for M.myotis are 76 % for fiber type II a, and 24 % for fiber type II b. Moreover the ratios of rapid fibers belonging to biceps brachii are more in M.schreibersi that of M.myotis, the ratio of type I fiber of serratus ventralis, biceps brachii and triceps brachii muscles in M.myotis are more then the ratios of type I fibers of the same muscles in M.schreibersi.Item Sıçanlarda oluşturulacak cerrahi yaralarda doku transglutaminazı etkisinin belirlenmesi(Fen Bilimleri Enstitüsü, 2018) Özgün, Orçun İşler; Cebesoy, Suna; Yazıhan, Nuray; Fen FakültesiTG2; hücre dışında yara iyileşmesi, anjiyojenez ve kemik onarımında önemli olan matriks stabilizasyonunda, hücre içinde ise apoptozis sırasında ve proteinlerin çapraz bağlanmasında rol oynayan transglutaminaz ailesinin en ilgi çekici üyelerinden biridir. Yaptığımız çalışmada; cerrahi kesi yöntemi ile yara oluşturulan sağlıklı ratlarda derialtı TG2 uygulamasının yara iyileşmesi üzerindeki etkinliği araştırılmıştır. Wistar Albino erkek ratlar; 8 adedi sağlıklı, 8 adedi kontrol cerrahi kesi grubu ve 8 adedi TG2 uygulanan cerrahi kesi grubu olmak üzere 3 gruba ayrılmıştır. Gruptaki kontrol cerrahi kesi grubu ve TG2 uygulanan cerrahi kesi grubu hayvanların sırt bölgelerine anestezi altında cerrahi kesi yara oluşturulmasının ardından sütur atılmıştır. Yara oluşumundan sonra ratların derisindeki fiziksel bazı değişiklikler 3., 5., 7., 10., 12. ve 15. günlerde fotoğraflanarak makroskobik olarak gözlenmiştir. 15. günde ratlar ötenazi edilerek süturlu bölgeleri çıkarılarak bir kısmı homojenizasyona kadar -80 °C'de muhafaza edilmiştir, bir kısmı ise % 4'lük paraformaldehidde tespit edilerek parafine gömülmüştür. Daha sonra -80 °C'de muhafaza edilen dokular homojenize edilip süpernatantları alınarak ELISA yöntemi kullanılarak Sitokrom c (apoptozis değerlendirmesi), kollajen-1, EGF (Epidermal Büyüme Faktörü), FGF (Fibroblast Büyüme Faktörü), VEGF (Vasküler Endotelyal Büyüme Faktörü), İnterlökin-8 ve -10 ölçümleri yapılmıştır. Parafine gömülen dokular ise mikrotomla kesitleri alınıp deparafinize edilerek Hematoksilen Eozin boyama yöntemi ve Masson'un Trikrom boyama kiti ile boyanıp mikroskopta histopatolojik olarak değerlendirilmiştir. Yara bölgesinin hem makroskobik olarak değerlendirildiğinde, hem ELISA sonuçları değerendirildiğinde, hem de mikroskobik olarak değerlendirildiğinde yara iyileşmesi açısından derialtı TG2 uygulanan grubun diğer gruplara göre daha iyi iyileşme gösterdiği görülmüştür. Sonuç olarak TG2'nin haricen olarak uygulandığında yara iyileşmesini hızlandırdığı görülmüştür. TG2; is one of the most interesting members of the transglutaminase family, which plays a role in matrix stabilization important for wound healing, angiogenesis and bone repair outside the cell, during apoptosis in the cell and for cross-linking proteins. In our work; the effectiveness of subcutaneous TG2 administration on wound healing was investigated in healthy rats that were wounded by surgical incision method. Male Wistar albino rats are divided three groups as 8 healthy tissue groups, 8 surgical incision groups and 8 TG2 treated surgical groups. Sutures were made to the dorsal regions of animals in the last two groups following surgical incision under anesthesia. Some physical changes in the skin of the rats after wound formation were macroscopically observed on the 3rd, 5th, 7th, 10th, 12th and 15th days by photography. At day 15 rats were euthanized and suture zones were removed. Some of the removed areas were kept at -80 °C until homogenization and the other part was fixed in paraformaldehyde at 4 % and embedded in paraffin. The tissues at -80 °C were homogenized and supernatants were obtained. And then Cytokrom c (apoptosis assessment), collagen-1, EGF (Epidermal Growth Factor), FGF (Fibroblast Growth Factor), VEGF (Vascular Endothelial Growth Factor), Interleukin-8 and -10 measurements were performed in the supernatants by ELISA method. The paraffin embedded tissues were sectioned by microtome and then they were deparaffinized. The deparaffinized tissues was stained with Hematoxylin Eosin staining method and Masson's Tricrom staining kit and histopathologically evaluated in the microscope. When the wound area was assessed macroscopically, when ELISA results were assessed and when microscopically assessed, the group treated with subcutaneous TG2 in terms of wound healing showed better recovery than the other groups. As a result, TG2 when applied externally had been seen to accelerate wound healing.Item The effect of melatonin on rat soleus muscle treated with carbon tetrachloride(Ankara Üniversitesi Fen Fakültesi, 2021) Cebesoy, Suna; Biyoloji; Fen FakültesiAntioxidants are known to restrains various tissue damage caused by the administration of carbon tetrachloride (CCl4). This study examined whether melatonin (MEL), a molecule known to have antioxidant properties, has a protective effect on the rat soleus muscle, where toxic damage is caused by the application of CCl4. In the study, eighteen albino-type male Wistar rats were used and divided into three groups as Control Group (group 1), CCl4 group (group 2) and CCl4 + MEL group (group 3). End of the 12 weeks, blood samples were taken as intracardiac from the rats under ketamine/rompun anesthesia, and the soleus muscles of the rats were removed. Tissue samples were subjected to routine preparation procedures for light microscopy. Sections 5 µm thick taken and stained with hematoxylin-eosin (HE) for histopathological examinations and Masson’s Trichrome stain for fibrosis formations. In conclusion, the CCl4 group displayed muscular hypertrophy, fiber orientation dysfunction and atrophy in some areas. In addition, fibrosis was spotted around the venous and nerve plexuses. In contrast to the CCl4 group, the melatonin group displayed no fibrosis and maintained tissue integrity. Therefore, when comparing CCl4+MEL and CCl4 groups, it was observed that melatonin had a stabilizing or even curative effect on the injuries.
