    Rekombinant insan insulin peptidinin bakteriyal ekspresyon sistemi kullanarak üretimi

    Insulin which is produced by pancreas with situated beta cells, has polypeptide structure. Insulin has 5400 daltons with a molecular weight and fifty one amino acids. Moreover the insulin hormone is the result of connected to each other by disulfide bonds of the A chains and B chains. This hormone plays a role in the regulation of blood sugar in human body. When the deterioration of the balance of blood glucose arises in the human body, it causes diabetes. This disease is seen among many people in the world, which is named such as Type 1 diabetes, Type 2 diabetes, gestational diabetes, and diabetes by specific causes. In this thesis the main goal is produced using bacterial production of human insulin producers systems for therapeutic, use recombinant DNA technology. Therefore, they are vectors encoding the entire sequence of the insulin gene have been created. The generated vectors used for the production bacteria system, transferred to the Escherichia coli, K12-JM109 and BL21 (DE3) cells and they are used for protein production. After that the protein production and purification have been optimized with the detailed study.
