Mesleki olarak arseniğe maruz kalan bireylerdeki glut1 gen polimorfizminin idrar arsenik düzeylerine etkisi

Choronic arsenic exposure cause to many disease and cancer. Some proteins and enzymes, which have a role in toxicokinetic of arsenic, effect pathogenesis of arsenic exposure- related diseases and cancer. Specially proteins and ezymes which responsible to exretion and acummulation of arsenic, have a big role in chronic effect formation. In this study, the aim was to determine the effect of polymorphism of gene Glut1 to urine arsenic level in 100 Turkish smelter workers. In this study, firstly, arsenic concentration of urine samples of 100 workers were measured by Graphite Furnace Atomic Absorption Spectroscopy (GFAAS) with Zeeman correction. In the second part of study, glut1 A2841T single nucleotide polymorphism was investigated by Polymerase Chain Reaction-Restriction Fragment Lenght Polymorphism (PCR-RFLP) method and the effects of these single nucleotide polymorphisms on urine arsenic levels were statistically analysed. As a result of statistical analysis, there was a statistically significant association between glut1 A2841T polymorphism and urine arsenic level. It was detected that the glut1 A2841T wild type genotypes have higher urine arsenic level than other genotypes. In conclusion, this study show that Glut1 A2841T single nucleotide polymorphism is associated with urine arsenic levels.
