ABSTRACT Master Thesis CALLUS INDUCTUON AND PLANT REGENERATION FOR DIRECT GENE TRANSFER IN WINTER TWO-ROWED AND SIX-ROWED BARLEY CULTIVARS Halide ACAR Ankara University Graduate School of Natural and Applied Sciences Department of Field Crops Supervisor: Prof. Dr. Günal AKBAY Two and six-rowed barley cultivars and lines adapted to Central Anatolia, were used to determine the callus induction and plant r ©feneration potentials for direct gene transfer was carried out at the experimental field and Biotechnology Laboratory of Field Crops Department of Agricultural Faculty of Ankara University. The culture responses were formed in MS and BS media from the seeds obtained from different research centers. In the formation of culture responses, mature and immature embryos were used, immature embryos were harvested from the crop of 1998-99 harvesting year. As the effect of the embryo type to culture response in different media were examined, in BS medium there is a difference between mature and immature embryos and no difference between plant regeneration and culture efficiency. And also the effect of MS and BS media to culture responses of mature and immature embryos were examined, there is no sound difference in the callus induction, plant regeneration and culture efficiency.
