New drug research is a long lasting and costly process. Therefore it is essential to eliminate the the unacceptably toxic or poorly active drug candidates at the earliest stage of the process and to continue to the further tests with the promising chemicals. In this context, cytotoxicity and genotoxicity test results provides unique data about the toxic concentrations of the chemicals at cellular and molecular (DNA) levels respectively. The aim of this master thesis is to determine the cytotoxic and genotoxic concentrations of the novel antibacterial and antifungal drug candidates synthesized by the pharmaceutical chemistry department.The novel drug candidates (13 chemicals) were subjected to neutral red uptake (NRU) cytotoxicity test in order to determine the IC50 values of the chemicals in HeLa cell lines. Sodium dodecyl sulphate (SDS) was used as positive control. According to the results of the cytotoxicity tests the IC50 values of 4 chemicals were statistically higher than the rest of the chemicals. The remaining 9 chemicals were considered as cytotoxic due to the quite low IC50 values. Therefore the alkaline comet assay was applied to 4 chemicals only. H2O2 was used as positive control. The results revealed that these 4 chemicals were not genotoxic at the concentrations lower than their IC50 values.According to these results the chemicals coded as 13, 20, 21 and C8 are the most promising novel antifungal/antibacterial drug candidates among the other nine chemicals.
