Çeltikte (Oryza sativa L.) endosperm destekli olgun embriyo kültüründe kallus oluşumu ve bitki rejenerasyonuna tohum iriliğinin etkisi

This study was carried out in University of Ankara, Faculty of Agriculture, Department of Field Crops, Biotechnology Laboratory during 2013-2014, mature embryos of 10 registered rice (Oryza Sativa L.) varieties (Aromatik-1, Çakmak, Durağan, Edirne, Efe, Gala, Halilbey, Neğiş, Osmancık-97, Şumnu) were used as plant material. These 10 rice varieties were seperated into 2 groups as small and large in in vitro conditions. Liquid MS medium including 8 mg/l 2,4-D (2,4-Dichlorophenoxyacetic acid) was used for callus induction. For plant regeneration, solid MS medium including 20 g/l sucrose and 7 g/l agar was used. In this study, callus induction, callus weight, regeneration capacity and culture efficiency parameters were determined. According to the results, 96,7-100,0 % callus induction, 0,468-0,837 gr callus weight, 42,0-86,7 % regeneration capacity and 40,0-86,7 % culture efficiency were obtained in large seeds. In small seeds, these parameters varied as 93,4-100,0 % in callus induction, 0,326-0,581 gr in callus weight, 36,0-63,3 % in regeneration capacity and 33,3-63,3 % in culture efficiency, respectively. It was established that; for rice, seed size is effective on plant regeneration and regeneration capacity is higher in larger seeds.
