PROGNOSTIC VALUE OF MMP-7-181 A/G SINGLE NUCLEOTID POLYMORPHISM AND HELICOBACTER PYLORI INFECTION AT GASTRIC CANCERGastric Cancer is widely seen all over the world, despite advances of surgery it is still a very serious health problem that causes high morbidity and mortality. Currently the main treatment method of this disease is surgery. 5-year disease-free survival of gastric cancer is very low so the researchers directed to research the factors affecting prognosis.In this study to investigate the effect of MMP-7 gene promoter region -181 A / G single nucleotide polymorphism in the development and prognosis of gastric cancer at Ankara University Faculty of Medicine Department of Medical Genetics, previously held " The impact of genomic changes on telomerase activity in gastric cancer" from the project titled the remaining cases to 34 cases of intestinal type gastric adenocarcinoma, Ankara University Faculty of Medicine, Department of Medical Genetics anonymously stored at -80 with the tumor and normal tissue samples and between 1/10/2008 and 1/10/2010 Department of Surgical Oncology, Ankara University Faculty of Medicine which is operated on 16 patients with a diagnosis of cancer of the stomach tumor and normal tissue samples were used.The study group consisted of 50 people, including 31 males and 19 females. The mean age was 58.5. As a result of the chain reaction-restriction fragment length polymorphism (PCR-RFLP) 50 patients with gastric cancer; 15 AA, 20 AG, 15 GG genotype was found. In the study of blood taken from volunteers; 25 AG, 9 GG and 16 AA genotype was found.G1 + G2 and G3 phases were compared according to A / G polymorphism. G allele frequency of stage G3 53.8% of patients with G1 + G2 phase was found 45.8%. According to this result than the frequency of G allele being considered to increase the G phase. However, p> 0.05 because of the provision of clinical compliance by increasing the number of subjects, this relationship can be predicted statistically significant.Patients' MMP-7 -181 A/ G polymorphism compared with the clinical status of individuals; G allele cariers were seen to have more larger tumor size and lymph node metastases. In the previous studies mRNA and MMP-7 levels found more higher in cancer tissues compared with the normal tissues but in our study there is no DNA level changes demonstrated to cause this condition. In our country and in Europe stomach cancer prognosis is worse than Japan because frequency of G allele is more than Japan.
