Geçici daldırma sistem biyorektörlerle su mercimeği (Lemna minor L.) bitkisinin in vitro çoğaltımı

Lemna minor L. rich in protein, found in large quantities in Turkey that help in reducing water pollution and play an important role in maintaining balance in aquaculture. This study reports role of Temporary immersion Bioreactors in micropropagation of this plant and effects of plant growth regulators on protein concentration in the plants. With this objective different variant of media with and without sucrose, varying pH and concentrations of BAP, kinetin, TDZ in medium were analyzed. These studies were carried out at 8 h dark and 16 h light photoperiod (150 µ mol photons m-2s-1 ) at 24±1°C. Maximum plant regeneration was obtained using 0.2 mg/l BAP in liquid MS medium without sucrose at pH 7.23. This medium regenerated 50.44 plants per explant. At the same time liquid MS medium containing 0.05 mg/l kinetin and 0.6 mg/l TDZ regenerated 57,823 and 50,74 plants per explant respectively. Protein measurement using Kjeldahl method showed 25.5% protein in the plant that increased to 29.18% using 0.5 mg/l BAP using hormone applications. It was concluded that the aim of the study in this thesis were fulfilled and positive effects of temporary immersion system bioreactors on plant multiplication were found.Key words: In vitro, Temporary immersion system bioreactors, Lemna minor L, micropropagation
