Yaban mersini ( Vaccinium myrtillus/Vaccinium corymbosum ) fenolik bileşiklerinin LC-MS/MS ile belirlenmesi
Abstract
In Turkey, there have been significant steps in cultivation of blueberry recently. This research has been devised to determine some phenolic compounds of blueberry in LC-MS/MS device. Analysis of variance shows important differences (p<0,005) between different varieties of blueberry in terms of pH, brix, titrattable acidity, color values of L*a*b, EC50 inhibition value, rutin, kaempferol, chlorogenic acid, epicatechin and quercetin. In this study, phenolic substances of blueberry were determined through LC-MS/MS method and the effects of parameters on the amount and distribution of these compounds were revealed. The main reason why we chose to use LC-MS/MS among other chromatographic techniques is the factor of repeatability and sensitivity. Antioxidant activities of these blueberries were also detected through DPPH method. According to our findings, titrattable acidity in blueberry was between 3,812 g/L and 17,094 g/L; brix was between 8,88 and 16,96; pH was between 2,715 and 3,18; the value of EC50 was 42,13 µL and 118,16 µL; L value was between 23,66 and 40,55; a value was between 10,90 and 30,93; b value was between 2,18 and 12; amount of rutin was between 0,0186 µg/mL and 0,968 µg/mL; amount of kaempferol was between 1,49 µg/mL and 26,60 µg/mL; amount of chlorogenic acid was between 0,0316 µg/mL and 8,050 µg/mL; amount of epicatechin was between 0,129 µg/mL and 0,142 µg/mL; amount of quercetin was between 0.0089 µg/mL and 0,208 µg/mL. The data obtained from this research have revealed that there are differences in the distribution of phenolic compounds and in % inhibitions among different varieties of blueberry. Chromatograms also prove our findings. In this study, the sample that provided the highest EC50 value was YM.11 and the sample that had the lowest EC50 value was YM.2. It is generally thought that differences in antioxidant activity result from the differences of compounds that have an antioxidant activity in phenolic substance concentrations of blueberries.
