In the previous reports, splenic macrophages of rats and mice have been extensively studied by their phagocytic uptake of carbon and thymus independent type-2 antigens, lysosomal digestions activities and staining properties with monoclonal antibodies. Macrophage subpopulations have been classified as the ones red pulp, in marginal zone and named as marginal metallophillics. The aim of this study is to show the phagocytic uptake of india ink material by macrophages located in the marginal zone of rat spleen and to trace the fate of necrobiotic and degenerative red blood cells during their lysosomal digestion in macrophage cytoplasm. The phagocytic activity of the marginal zone macrophages were low in relation to the carbon particles. Same cells showed moderate positive reaction about acid phosphatase. Non-specific esterase reaction did not give positive results in macrophage cytoplasms as granules and vacuoles seen free from precipitations. As for the marginal metallophilis, they displayed few phagocytic reaction for carbon particle and intensive precipitations by acid phosphatase enzyme. They have also been stained when applied Pearl's iron stain method. Macrophages of red pulp area displayed highly strong phagocytic activity for carbon particles, intensive positive acid phosphatase precipitations and moderate non specific esterase precipitations. They were also strongly stained by Pearl's iron method. They had granules and vacuoles in their cytoplasms as observed in semi-thin sections. In this study fine morphological properties and abilities of phagocytic uptake of the marginal zone macrophages related to the carbon particles were studied. They were previously evaluated as the cells utilizing thymus independent type-2 antigens and transferring it to the B lymphocytes of marginal zone as a stimulator. This study confirmed the result above. It has been decided that the principal phagocytic cells of the spleen should be the macrophages of red pulp zone.
