In this study, the changes in high serotonin and noradrenaline levelsthat are expected to happen in blood and the effects on zinc and iron levelsin tissues of liver, kidney and adrenal glands are examined, following the highdose application of sibutramine hydrochloride, drug substance of a widelyused antiobesity drug, on rats.The study was conducted at Gazi University Experimental ResearchCenter. In this study, serotonin and noradrenaline levels in blood samplesand zinc and iron levels in tissues of liver, kidney and adrenal glands of 15rats were compared, 8 in control group, 7 in sibutramine application group.Serotonin and noradrenaline levels were determined by HPLC ECD and themetal analysis in the tissues were determined by flame AAS method. Bloodand tissues of control group rats were taken without giving any substanceexcept for anaesthesia. Sibutramine application group was injected 20 mgsibutramine per kg and their blood and tissues were taken 72 hours later,blood samples were sent to Düzen Laboratory and tissues were sent to theInstitute of Forensic Medicine.The statistical analysis of the results was evaluated with the SPSS16.0 program. As a result of the study, mean serum serotonin levelsmeasured in control group and sibutramine application group weredetermined as 470,625 ?g /L, 1122,857 ?g /L, respectively (p<0.05).Metal levels in liver tissue were compared statistically in control groupand sibutramine application group and no significance was found. When78control and sibutramine application groups were compared in serotoninvalues, the values were found as 1122,85± 40,70 in sibutramine applicationgroup and 470,62± 43,65 in control group, these values were found to bestatistically significant.Iron values in kidney tissue in control and sibutramine applicationgroups were determined as 78,22 ?g /L, 108,27?g /L, respectively (p<0.05).Zinc values in kidney tissue in control and sibutramine groups weredetermined as 346,94?g /L, 446,48?g /L, respectively (p<0.05).Iron values in adrenal glands tissue in control and sibutramineapplication groups were determined as 104,18 ?g /L, 174,00 ?g /L,respectively (p<0.05). Zinc values in adrenal glands tissue in control andsibutramine application groups were determined as 0,08 ?g /L and 2,84 ?g/L, respectively (p<0.05).It was recorded that rats in the group that sibutramine was applicated,had loss of appetite and significant weight loss, and their food consumptiondecreased in half.
