Maedi-Visna enfeksiyonunun tanısında PCR tekniğinin kullanımı ve yerel virusların moleküler karakterizasyonu

In this study, it was aimed that, the use of ELISA and PCR techniques for diagnosis of MVV infection in sheep blood samples obtained from different regions of Turkey and molecular characterization of fields viruses. 821 blood samples were tested for antibody against to MVV and investigated to viral nucleic acide using primers spesific to LTR region of viral genome by PCR. SCP and Foetal Sheep Lung cells were used for virus isolation and idendification.Out of 561 (68,3%) blood samples were found as positive by ELISA and/or PCR respectively. Also it is found that posivity rate of the infection increased according to age and differed by breed. The possivity rate of Karayaka breed are implicated less than Akkaraman, Merinos and mix breeds. 32 field viruses, were found by PCR as positive, analysed their LTR regions and compared with each other and other referenced LTR sequences on GenBank. However the field viruses have differences each other, are implicated they closed with American and Greece strains.When compare to the results of ELISA and PCR in this study, the posivity rate of ELISA were found higher than PCR and the posivity rate of PCR in young animals is higher than other ages. As conclusion, is suggest that should be use of ELISA systems in eradication of MVV infection and use of PCR tecniques for negatif samples by ELISA esspecially early stages of the infection.
