Çeşitli bitkisel ekstraktlar ve kombinasyonlarının köftede antimikrobiyel etkisinin araştırılması

Bu çalışmada, bazı bitki materyallerinin ve kombinasyonlarının E. coli O157:H7, S. Enteritidis, E. coli Biyotip 1, L. monocytogenes ve S. aureus'a karşı antimikrobiyel aktiviteleri disk difüzyon yöntemiyle araştırılmıştır. Zeytin yaprağı ekstraktı, propolis, greyfurt uçucu yağı, portakal kabuğu uçucu yağı ve biberiye uçucu yağında ve bu materyallerin kombinasyonlarındaki antimikrobiyel aktiviteler incelendiğinde Gram pozitif bakterilerden özellikle S. aureus'a karşı yüksek inhibisyon zon çapları gözlemlenmiştir. Literatürde yer alan, antimikrobiyel etkinin genel anlamda Gram pozitif bakterilere karşı daha etkili olduğu bulgusu bir kez daha kanıtlanmıştır. Her ne kadar arı ürünü olsa da materyalin ağırlıklı olarak bitki kökenli olması nedeniyle bu çalışmada propolis de kullanılmıştır. Patojenlere karşı en yüksek inhibisyon zonu oluşturan materyaller greyfurt uçucu yağı ve biberiye uçucu yağı olmuştur. Bitki materyallerinin 2'li kombinasyonlarının en yüksek antimikrobiyel etki gösterdiği Gram negatif bakteri, S. Enteritidis olurken 3'lü kombinasyonların en yüksek antimikrobiyel etki gösterdiği Gram negatif bakteri ise E. coli O157:H7 olmuştur. Bu çalışmada tek başına antimikrobiyel aktiviteye sahip olmayan materyallerin farklı sayıda kombinasyonlarının sinerjik etki oluşturabileceği ve bazı kombinasyonlarda da birbirlerini baskılayarak aktiviteyi düşürebildikleri gözlemlenmiştir. Yapılan toplam fenolik miktarı analizi sonucunda en çok toplam fenolik miktarı greyfurt uçucu yağında gözlemlenmiştir. Çalışmada kullanılan bitki materyallerinin antimikrobiyel aktivite sonuçları ile toplam fenolik miktarlarının sonuçları birbiri ile paralellik göstermektedir. Çalışma için seçilen 5 bitki materyali ve materyallerin 3'lü kombinasyonlarından 2 tanesi duyusal analiz amacıyla köfteye eklenmiştir. Panelistlerin renk, görünüş, koku, tat, tekstür ve genel beğeni değerlendirmeleri sonucunda kontrol köftesine en yakın kriterleri zeytin yaprağı ekstraktı içeren köftenin gösterdiği ve 3'lü kombinasyonların en düşük beğeni puanlarını aldığı tespit edilmiştir.
In this study, antimicrobial activities of some plant materials and combinations against E. coli O157:H7, S. Enteritidis, E. coli Biotype 1, L. monocytogenes and S. aureus were investigated by disk diffusion method. When the antimicrobial activities of olive leaf extract, propolis, grapefruit essential oil, orange peel essential oil and rosemary essential oil and combinations of these materials were examined, high inhibition zone diameters were observed against Gram positive bacteria, especially S. aureus. The finding in the literature that the antimicrobial effect is generally more effective against Gram positive bacteria has been proven once again. Although it is a bee product, propolis was also used in this study because the material is mainly of plant origin. The materials forming the highest zone of inhibition against pathogens were grapefruit essential oil and rosemary essential oil. The Gram negative bacterium with the highest antimicrobial effect of the 2-in-1 combinations of plant materials was S. Enteritidis, while the Gram negative bacterium with the highest antimicrobial effect in the triple combinations was E. coli O157:H7. In this study, it was observed that different combinations of materials that do not have antimicrobial activity alone can create a synergistic effect, and in some combinations, they can reduce the activity by suppressing each other. As a result of the analysis of total phenolic content, the most total phenolic content was observed in grapefruit essential oil. The results of the antimicrobial activity and total phenolic amounts of the plant materials used in the study are in parallel with each other. 5 plant materials selected for the study and 2 of their triple combinations were added to the meatballs for sensory analysis. As a result of the panelists' evaluations of color, appearance, smell, taste, texture and general taste, it was determined that the closest features to the control meatballs were shown by the meatballs containing olive leaf extract and the combinations of 3 received the lowest scores.
