Asetilkolin tayini için nanomateryal temelli elektrokimyasal biyosensörlerin geliştirilmesi

Bu çalışmada Alzeheimer, parkinson ve şizofreni hastalıklarının teşhisinde önemli bir belirteç olan asetilkolini tespit etmek için hızlı ve güvenilir bir biyosensör geliştirilmiştir. Farklı nanoyapılar ile modifiye edilen camsı karbon elektrotlar ile hazırlanan biyosensörler asetilkolin tayini için kullanılmıştır. Geliştirilen bu biyosensör ile asetilkolin tayini enzim-substrat ilişkisine dayanarak yapılmıştır. Günümüzde yaygın olarak asetilkolin tayininde LC (sıvı kromatografisi) ve MS (kütle spektroskopisi) kullanılmaktadır. Bu yöntemlerin maliyetinin yüksek olması, uzmanlık gerektirmesi, çeşitli ön işlem ve yıkama aşamalarının zaman alması ve aynı anda çoklu analiz yapamaması gibi dezavantajları bulunmaktadır. Bu sebepleri göz önünde bulundurarak maliyeti düşük, uygulama basamakları daha az, hızlı ve güvenilir sonuç verebilecek bir nanomateryal temelli elektrokimyasal biyosensör geliştirilmiştir. Nanopartiküllerin hızlı elektron transferi, etkin geniş yüzey alanı, yüksek yüzey enerjisi, protein-metal partikülleri yaklaştırma özelliği bulunmaktadır. Ayrıca elektroanalitik işlemler için hassas, kolay modifiye edilebilir, kararlı bir elektrot yüzeyi oluştururlar. Asetilkolin tayini için hazırlanan bu amperometrik biyosensörde çalışma elektrodu olarak camsı karbon elektrot ve modifiye edici olarak kitosan, çok duvarlı karbon nanotüp (MWCNT) ve Fe3O4 nanopartiküller kullanılmıştır. Hazırlanan biyosensör serum örneklerinde asetilkolin seviyelerini ölçmek için kullanılmıştır.
In this study, a fast and reliable biosensor was developed to detect acetylcholine which is an important marker in the diagnosis of Alzeheimer, Parkinson's and schizophrenia diseases. Biosensors prepared with glassy carbon electrodes modified with different nanostructures were used for the determination of acetylcholine. This biosensor was developed based on the enzyme-substrate relationship with acetylcholine. LC (liquid chromatography) and MS (mass spectroscopy) are widely used for the determination of acetylcholine. These methods have some disadvantages such as high cost, expertise requirement, various pre-treatment and washing stages take time, and multiple analysis can not be performed at the same time. Considering these reasons, a nanomaterial-based electrochemical biosensor has been developed which can provide low cost, low application steps, fast and reliable results. Nanoparticles have fast electron transfer, effective large surface area, high surface energy, ability to approximate protein-metal particles. They also form a precise, easily modifiable, stable electrode surface for electroanalytical processes. The glassy carbon electrodes were used as working electrode and the chitosan, multiwall carbon nanotubes (MWCNTs) and Fe3O4 nanoparticles were selected as modifiers for the preparation of acetylcholine biosensor. The fabricated biosensor was used to determine acetyhlcholine levels in serum samples.
