dc.description.abstractBu tez çalışmasının ilk aşamasında, insan somatik dermal fibroblast hücreleri, insan multipotent kemik iliği mezenkimal kök hücreleri ve insan uyarılmış pluripotent kök hücreleri kültürde çoğaltılıp moleküler ve immünfenotipik yöntemlerle tanımlandı. Daha sonra, bu üç farklı potansiyele sahip hücre tipini in vitro koşullarda deselülerize ekstraselüler matriks (ECM) elde etmek amacıyla deselülerizasyon protokolleri optimize edildi. Protokollerin etkinliği histokimyasal ve spektrofotometrik DNA içeriği analizleri ile belirlendi. Elde edilen üç farklı tipteki deselülerize ECM'nin morfolojik özellikleri, büyüme faktörleri bileşimleri, glikozaminoglikan bileşimleri ve protein içerikleri ayrıntılı olarak incelendi. Dermal fibroblast hücrelerinin ECM'ler üzerine tohumlanması sonrasında morfolojik davranışları, canlılıkları ve çoğalma yeteneklerindeki değişimler ele alındı. Son aşamada, elde edilen farklı tipteki ECM'leri içeren üç-boyutlu (3B) doku iskeleleri elektroeğirme tekniği kullanılarak hazırlanıp karakterize edildi. Geliştirilen 3B nanofiberlerin hücreler üzerindeki etkileri in vitro koşullarda eş-zamanlı sitotoksisite ve elektron mikroskobu analizleri ile incelendi. Elde edilen sonuçlarda farklı potense sahip hücrelerden ECM eldesine yönelik NH4OH/Triton X-100'ün kullanılabileceği belirlendi. Bu sayede kültürde çoğaltılan hücrelerin çekirdeklerinin ve bulundurdukları DNA'nın önemli düzeyde uzaklaştırılabildiği ve aynı zamanda sGAG, büyüme faktörleri ve ECM proteinlerinin deselülerize hücre kültürü temelli ECM'lerde korunabildiği belirlendi. Farklı potensteki hücrelerden toplanan ECM'lerin in vitro kültür çalışmalarında ve 3B doku iskeleleri için biyoaktif bileşen olarak kullanılabileceği ve ayrıca somatik hücre davranışlarının deselülerize ECM'ler ile düzenlenebileceği bu tez çalışması ile belirlendi. In the first part of this thesis study, human somatic dermal fibroblasts, multipotent bone marrow mesenchymal stem cells and induced pluripotent stem cells were cultured and identified by molecular and immunophenotypical methods. Decellularization protocols were optimized in order to obtain the extracellular matrix (ECM) in vitro from these three different cell types with different potencies. The efficiencies of the protocols were determined by histochemistry and spectrophotometric DNA content analyses. The three different types of the decellularized ECMs were examined for their morphological characteristics, growth factor contents, glycosaminoglycan contents and protein contents. Changes in morphological behaviour, cell viability and the proliferation capacity of dermal fibroblasts seeded on different types of ECMs were determined. Finally, three-dimensional (3D) scaffolds containing three different types of ECMs were fabricated by the electrospinning technique and then were characterized. The in vitro effects of the 3D nanofibers on the cells were examined by simultaneous cytotoxicity and electron microscopy analyses in detail. Findings showed that NH4OH/Triton X-100 could be efficiently used for the isolation of ECMs from the cells with different potencies. It was found that the DNA content and cell nuclei could be removed from the cells, while at the same time preserving the sGAGs, growth factors and ECM proteins in the cell culture-derived ECMs to a significant extent by decellularization. This thesis study clearly showed that the ECMs collected from different types of cells could be used as a bioactive component in in vitro culture studies, as well as in 3D tissue scaffold applications, and that somatic cell behaviour could be regulated by decellularized ECMs.tr_TR
