dc.description.abstractTürkiye'de farklı şarap bölgelerinde izole edilen 3 adet Saccharomyces cerevisiae suşlarının ve bir adet referans suşun farklı stres faktörleri karşısında davranışları incelenmiştir. Her bir stres faktörünün farklı düzeyleri suşlara ayrı ayrı uygulanmış, süreye bağlı olarak şuşların maya sayıları ve hücresel trehaloz düzeyleri belirlenmiştir. Sıcaklık stresi uygulamaları içerisinde, 2, 7, 47 ve S122 kodlu suşların inaktivasyonunu en fazla etkileyen uygulamanın 55 °C olduğu, YPG besiyeri ortamında 45. dakikada suşların inaktivasyon oranlarının sırasıyla 5,20; 4,68; 3,84 ve 4,22 log olduğu belirlenmiştir. Etanol uygulamaları içerisinde, en yüksek inaktivasyonun % 14 etanol uygulamasına ait olduğu, ait 2, 7, 47 ve S122 kodlu suşlar için inaktivasyon oranlarının sırasıyla 4,54; 4,93; 4,38 ve 4,64 log olduğu tespit edilmiştir. Stres uygulamalarının şiddeti ve uygulanma süresi arttıkça, suşların trehaloz düzeylerinin arttığı, en yüksek hücresel trehaloz düzeylerinin sıcaklık stresinde 45 ve 50 °C, etanol stresinde % 14 konsantrasyonunda elde edildiği ve bu streslerde suşların hücresel trehaloz miktarının süreye bağlı olarak fazla değişmediği bulgularına ulaşılmıştır. Farklı ön işlemler sonrası, fermantasyon ortamına bırakılan suşların tükettikleri glikoz ve fruktoz miktarları üzerine suş farkının, uygulama farkının ve sürenin etkisinin önemli olduğu (p<0,05), etanol üretimi üzerine uygulama farkı etkisinin önemli olduğu ve bu farkın etanol ön uygulaması sonunda etanol düzeyinde gözlemlenen artıştan kaynaklandığı belirlenmiştir. Ön etanol uygulaması sonrası, 48 saat fermantasyon sonunda suşların ürettikleri etanol miktarı sırasıyla; 76,50; 70,45; 87,30 ve 85,30 g/L olarak tespit edilmiştir. Sigmoidal fonksiyonda ile tüm suşların uygulamalar sonrası elde edilen R2 değerinin en düşük 0,964 olduğu, modelin uygulamalar sonrası fruktoz kullanımını çok iyi tanımladığı görülmüştür. The behaviour of 3 Saccharomyces cerevisiae strain, isolated from different wine region in Turkey and one reference strain against stress were investigated. Different levels of each stress factor were applied to the strains, separately and yeast counts and intracellular trehalose levels of the strains at each stress level were determined. In the temperature stress applications, the most effective applicatiom was found at 55 °C with the inactivation rates 5.20, 4.68, 3.84 and 4.22 log for 2, 7, 47 and S122 coded strains, respectively. Among the ethanol applications, the highest inactivation belongs to 14 % ethanol application and the inactivation rates for the 2, 7, 47 and S122 coded strains were 4.54, 4.93, 4.38 and 4.64 log, respectively. As the levels and duration of stress applications increased, production of intracellular trehalose levels among all strains increased, the highest intracelluler trehalose levels were obtained at 45 and 50 °C in temperature stresses, similarity 14% concentration in ethanol stresses also increased the amount of intracellular trehalose level without change much regarding to the duration. After the different pre-treatments, it was observed that the effect of strain differences, application differences and durations on the amount of consumed glucose and fructose by the strains was not significantly important (p>0.05), but the effect of application difference on ethanol production was significantly important (p<0.05). It was observed that the difference was due to the increased after ethanol pre treatment. After the pre-ethanol applications, the amount of ethanol produced by the strains at the end of fermentation for 48 hours, 76.50; 70.45; 87.30 and 85.30 g/L, respectively.tr_TR
