Nohutlarda kök çürüklüğüne sebep olan funguslar arasındaki genetik farklılığın moleküler yöntemlerle incelenmesi

Ülkemizdeki nohut üretimi farklı biotik ve abiotik streslerden dolayı istenilen düzeyde olmamaktadır. Biotik hastalık etmenleri arasında ise basta Fusarium oxysporum f. sp. ciceris' in sebep olduğu Fusarium solgunluğu olmak üzere diğer kök patojenleri önemli bir yer tutmaktadır. Bu sebeple Türkiye'nin 15 farklı ilinde 2001-2002 yıllarında yapılan survey sonucunda, nohutlarda solgunluk ve kök çürüklüğüne sebep olduğu tespit edilen fungus izolatları patojenisite testi, Random amplified polymorphic DNA (RAPD) ve Inter simple sequence repeats (ISSR) yöntemleri kullanılarak karakterize edilmiştir. Yapılan survey sonucunda Fusarium oxysporum, F. solani, F. equiseti, F. semitectum, F. acuminatum, Macrophomina phaseolina ve Rhizoctonia solani nohutlarda solgunluk ve kök çürüklüğüne neden olan hastalık etmenleri olarak saptanmıştır. Elde edilen kök patojenleri arasında en yaygın patojenin F. oxysporum olduğu ve bunu F. solani ve M. phaseolina' nın takip ettiği görülmüştür. Hassas nohut çeşitleri kullanılarak toplam 144 izolatın virulenslikleri belirlenmiş ve bu fungus türlerindeki intra ve inter spesifik polimorfizimler 30 RAPD ve 20 ISSR primeri kullanılarak incelenmiştir. Elde edilen verilerin Dice' ın benzerlik katsayısı kullanılarak yapılan UPGMA (Unweighed Pairgroup Method with Arithmetic Average) cluster analizi sonucunda 74 F. oxysporum izolatı arasında genel olarak 3 ana grubun bulunduğu 25 F. solani izolatının ise 2 farklı gruba ayrıldığı tespit edilmiştir. Ayrıca nohutlarda hastalığa sebep olan diğer patojenler arasındaki genetik varyasyon her iki yöntem kullanılarak araştırılmış ve cluster analizi ile birbirlerinden ayrı olarak gruplandırılmışlardır. Moleküler yöntemler kullanılarak yapılan analizler sonucunda morfolojik kriterler kullanılarak ayırt edilemeyen izolatlar dendogramdaki dağılımlarına göre yeniden sınıflandırılmışlardır. RAPD, ISSR ve RAPD+ISSR verilerinin kombine edilmesiyle elde edilen dendogramlar arasında ise yüksek bir korelasyonun olduğu ve her iki yöntemin fungus populasyonları arasındaki genetik varyasyonu incelemek için oldukça faydalı olduğu tespit edilmiştir.Abstract Chickpea production is not at the desired level because of biotic and abiotic factors in Turkey. Fusarium wilt, caused by Fusarium oxysporum f. sp. ciceris and other root rot pathogens are important biotic disease agents. In this study, fifteen provinces of Turkey were surveyed for fungal pathogens, causing wilt and root rot on chickpea in 2001 and 2002. The fungal species obtained were characterized with pathogenicity test, Random amplified polymorphic DNA (RAPD) and Inter simple sequence repeats (ISSR) assays. As a result of the survey, Fusarium oxysporum, F. solani, F. semitectum, F. acuminatum, F. equiseti, Macrophomina phaseolina and Rhizoctonia solani were detected as pathogens, causing wilt and root rot on chickpea. F. oxysporum was the most widespread pathogen among root rot pathogens, followed by F. solani and M. phaseolina. The virulence spectrum of a total 144 isolates was determined on susceptible chickpea cultivars, and intra and inter specific polymorphisms among these fungal species were investigated using 30 RAPD and 20 ISSR primers. UPGMA (Unweighed Pairgroup Method with Arithmetic Average) cluster analysis of the data from DNA amplification of RAPD and ISSR using Dice?s coefficient showed 3 major groups among 74 isolates of F. oxysporum and also, clustered 25 isolates of F. solani into 2 different groups. In addition, genetic variation among other pathogens causing disease on chickpea plants was analyzed using both techniques and grouped separately from each other with cluster analysis. The isolates that were not easily separable using morphological characteristics were classified again according to their branching in dendrograms. The results showed that there was a high correlation between dendrograms obtained from the data of RAPD, ISSR and combined RAPD+ISSR and both techniques were useful to study genetic variation among fungal populations
