Koyunlarda corynebacterium pseudotuberculosis'in ELISA ve dot-blot ELISA ile teşhisi

Koyunlarda Corynebacterium pseudotuberculosis' in ELISA ve Dot-blot ELISA ile Teşhisi Bu çalışmada, koyun kan serumlarında C. pseudotuberculosis antikorlarının saptanması için; ekzotoksin ELISA, sonike ELISA ve dot-blot ELISA teknikleri standardize edilerek, infeksiyonun rutin teşhisinde ve bir tarama testi olarak da laboratuvar çalışmalarında kullanılabilirliklerinin ortaya konulması amaçlandı. Araştırmada, Tarım İşletmeleri Genel Müdürlüğû'ne bağlı Ceylanpınar, Tahirova, Altınova, Gözlü ve Polatlı Tanm İşletmelerinden sağlanan, 100 adeti aşılı hayvanlardan olmak üzere toplam 471 adet koyun kan serumu incelendi. ELISA antijenleri A. Ü. Veteriner Fakültesi Mikrobiyoloji Anabilim Dalı Kültür Koleksiyonu'ndan sağlanan C. pseudotuberculosis Pl-4 susundan hazırlandı. Çalışmada, toplam 471 serum örneğinden 172 (% 36.5)'si ekzotoksin ELISA ile pozitif; 299 (% 63.5)'u negatif; 119 (% 25.2)'u sonike ELISA ile pozitif, 352 (% 74.8)'si negatif; 181 (% 38.5)'i dot-blot ELISA ile pozitif, 290 (% 61.5)'ı ise negatif olarak saptandı. Aşısız hayvanlara ait serumlardan ekzotoksin ELISA ile 96 (% 25.8)'sı pozitif, 275 (% 74.1)'i negatif, sonike ELISA ile 82 (% 22.2)'si pozitif, 289 (% 77.8)'u negatif bulunurken; dot-blot ELISA ile 122 (% 32.8)'si pozitif, 249 (% 67.2)'u negatif bulundu. Aşılı koyun serumlarından 76 (% 76)'sı ekzotoksin ELISA 37 (% 37)'si sonike ELISA ve 59 (% 59)'u da dot-blot ELISA ile pozitif olarak değerlendirildi. Test serumları toplam olarak değerlendirildiğinde, ekzotoksin ELISA'nın sensitivitesi % 82.9, spesifitesi % 65.9; sonike ELISA'nın sensitivitesi % 80.0, spesifitesi % 64.2; her iki test birlikte değerlendirildiğinde ise sensitivite % 93.3, spesifite de % 70.8 olarak tespit edildi. Dot-blot ELISA'mn sensitivitesinin % 83.3, spesifitesinin ise % 65.5 olduğu saptandı. Ekzotoksin ve sonike ELISA tekniklerinin birlikte kullanılması durumunda, testlerin duyarlılık ve özgüllüklerinin daha yüksek olduğu ortaya konuldu. Ayrıca ekzotoksin ELISA'nın, sonike ve dot-blot ELISA tekniklerinden daha güvenilir sonuçlar verdiği görüldü. Bu araştırmada elde edilen sonuçlar, koyunların kazeöz lenfadenitisinde ekzotoksin ELISA sonike ELISA ve dot-blot ELISA tekniklerinin, hastalığın rutin teşhisinde ve bir tarama testi olarak da laboratuvar çalışmalarında uygulanabileceğini gösterdi. AbstractDiagnosis of Ovine Corynebacterium pseudotuberculosis by ELISA and Dot-blot ELISA The aim of this study was to standardize the three techniques designated as exotoxin ELISA, sonicated ELISA and dot-blot ELISA in the detection of antibodies to Corynebacterium pseudotuberculosis in sheep sera and to evaluate the usefulness of these techniques in the routine diagnosis and screening of C. pseudotuberculosis infection. A total of 471 sheep sera, 100 of which were from vaccinated animals, obtained from Ceylanpinar, Tahirova, Altmova, Gözlü and Polatlı agriculture administrations subordinating Agriculture Administrations Directory were examined. ELISA antigens were prepared from C. pseudotuberculosis Pl-4 strain provided from the culture collection of the Department of Microbiology of the Faculty of Veterinary Medicine, University of Ankara. Among 471 serum samples, 172 (36.5 %), 119 (25.2 %) and 181 (38.5 %) sera were positive and 299 (63.5 %), 352 (74.8 %) and 290 (61.5 %) sera were negative by exotoxin ELISA, sonicated ELISA and dot-blot ELISA techniques, respectively. In the examination of serum samples obtained from non-vaccinated animals, 96 (25.8 %), 82 (22.2 %) and 122 (32.8 %) were positive and 275 (74.1 %), 289 (77.8 %) and 249 (67.2 %) serum samples were negative by the same techniques, respectively. Concerning the sera taken from vaccinated sheep, 76 (76 %), 37 (37 %) and 59 (59 %) were found to be positive by exotoxin ELISA, sonicated ELISA and dot-blot ELISA, respectively. When the evaluation of total test sera was made, sensitivities and specificities of the three techniques were found as follows, respectively: Exotoxin ELISA, 82.9 %, 65.9 %; sonicated ELISA, 80.0 %, 64.2 %; dot-blot ELISA, 83.3 %, 66.5 %. When the exotoxin and sonicated ELISA were evaluated together, sensitivity and specificity were estimated as 93.3 % and 70.8 %, respectively. According to the results of these evaluations, when exotoxin and sonicated ELISA techniques were performed together, sensitivities and specificities of the tests were found to be higher and exotoxin ELISA was observed to give more reliable results than the other two techniques. The results obtained in this research showed that, exotoxin ELISA, sonicated ELISA and dot-blot ELISA techniques could be performed as screening tests for the routine diagnosis of the caseous lymphadenitis disease of sheep.
