Abstract In this study, the cytological, anatomical, external morphological, physiological and palynological properties of diploid (2n=14) and tetraploid (2n=28) Secale cereale L. were comparatively investigated using light and electron microscopy. The plant height are higher and leaf breadth and lengths are larger in tetraploid S. cereale L. compared with those of diploids. Also the leaves and stems of tetraploids are thicker and their seeds are larger. The lengths and breadth of their leaves epidermal cells and stomata and volumes of their mesophyll cells are larger. The mesophyll cell organelles of tetraploid plants are richer than diploid plants with respect to their numbers and size. The number of grana and grana lamellae and stroma lamellae and starch grains in chloroplast are greater in tetraploids. Also the number of chloroplasts are higher. That is why, the total chlorophyll, chlorophyll a and chlorophyll b were found to be higher in tetraploid plants. The pollen grains of the tetraploids are larger and its pore number is higher than its diploid counterparts.
