Bazı termofilik Bacillus türlerinin termostabil α-glukozidaz üretim kapasiteleri ve enzim/lerin kısmi karakterizasyonu

α-Glukozidaz üreticisi termofilik Bacillus spp. izolatlarını belirlemek amacıyla Aydın, Ankara, Denizli, İzmir, Manisa ve Nevşehir illerinde bulunan 46 farklı s ıcak su kaynağından toplanan 191 adet örnekden koloni morfolojileri yönünden farklılık gösteren toplam 451 adet termofilik Bacillus izolatı elde edilmiştir. Yüksek amilaz aktivitesine sahip 67 adet termofilik Bacillus izolatı seçilerek izolatların hücre içi ve hücre dışı α- glukozidaz üretim kapasiteleri, pNPG substratı kullanılarak belirlenmiştir. İzolatların α- glukozidaz üretme kapasiteleri, toplam enzim miktarları baz alınarak belirlenmiş ve izolatların pelet yaş ağırlığına göre toplam α-glukozidaz miktarları 77.18-0.001 U/ml/g arasında bulunmuştur. Yüksek enzim üretme kapasitesine sahip Bacillus sp. A333 (48.64 U/ml/g), A343 (10.79 U/ml/g), E134 (32.09 U/ml/g), F84a (36.64 U/ml/g) ve F84b (77.18 U/ml/g) izolatlarının 48 saatlik zamana bağlı α-glukozidaz üretimleri, standart olarak kullanılan G. stearothermophilus ATCC 12980 suşu (34.26 U/ml/g) ile karşılaştırılarak belirlenmiştir. Hücre içindeki maksimum enzim üretimi zamanı 4. saat (A343, E134), 8. saat (F84a, F84b) ve 12. saat (A333) olarak belirlenmiş olup izolatlara göre değişiklik göstermiştir. Bacillus sp. A333 izolatı hücre dışı, Bacillus sp. A343 izolat ise hücre içi α-glukozidaz üreticisi olarak seçilmiş ve kısmen saflaştırılan enzimlerin %10’luk N-PAGE ve 4-MUG substratı ile yapılan aktivite analizi sonuçları, A333 ve A343 izolatlarında ayırıcı birer enzim bandı yanında standart suşlarda da elektroforetik mobiliteleri aynı olan ortak bir enzim bandı olduğunu göstermiştir. α-Glukozidaz enzimi daha sonra N-PAG’deki bu bantlardan ekstrakte edilerek %10’luk SDS-PAGE’de ayrılmış ve Bacillus sp. A333 ile A343 enzimlerinin, molekül ağırlıkları 44 ve 60 kDa civarında ikişer adet farklı proteine sahip oldukları belirlenmiştir. A333 α-glukozidazı optimum pH’ı 6.8, optimum aktivite sıcaklığı 60ºC ve pNPG substratına karşı Km değeri 1.38 mM olarak bulunurken, bu değerler A343 enzimi için sırasıyla 7.0, 65ºC ve 0.85 mM olarak belirlenmiştir. İki izolata ait enzimlerin 20 farklı substrata olan spesifikliği ve ince tabaka kromatografisi sonuçları A333 α-glukozidazının yüksek derecede transglukozilasyon aktivitesine, A343 enziminin ise geniş substrat spesifikliğine sahip olduğunu göstermiştir. Bunun yanısıra, Bacillus sp. A333 α-glukozidazının, A343 enzimine göre daha geniş bir pH ve sıcaklık aralığında stabilite gösterdiği ve her iki enzimin de test esilen inhibitörlerin çoğuna dirençli olduğu bulunmuştur. Yapılan bu çalışma ile biyoteknolojik uygulamalarda kullanım potansiyeline sahip yeni termostabil ekzo-α-1,4-glukozidaz üreticisi iki Bacillus sp. izolatı belirlenmiştir.AbstractA total of 191 samples were collected from 46 different hot springs in Aydın, Ankara, Denizli, İzmir, Manisa and Nevşehir cities in order to determine the α-glucosidase producer thermophilic Bacillus spp. From these samples 451 thermophilic Bacillus isolates having different colony morphology were obtained and screened for amylase activity. Sixtyseven of 451 thermophilic Bacillus isolates which had high amylase activity were selected to determine the intracellular and extracellular α-glucosidase production capacities by using pNPG substrate. α-Glucosidase production capacities of the isolates were designated basing on their total enzyme amount per pellet wet weight, and were found between 77.18-0.001 U/ml/g. Time course of high enzyme producers Bacillus sp. A333 (48.64 U/ml/g), A343 (10.79 U/ml/g), E134 (32.09 U/ml/g), F84a (36.64 U/ml/g) and F84b (77.18 U/ml/g) were determined by comparing with standard strain G. stearothermophilus ATCC 12980 (34.26 U/ml/g) for 48 hours. The maximum intracellular enzyme production time was found as 4 hours (A343, E134), 8 hours (F84a, F84b) and 12 hours (A333) which showed variations according to the isolates. Bacillus sp. A333 and A343 were selected as an extracellular and intracellular α- glucosidase producers, respectively. After partial purification of α-glucosidases from two isolates, 10% N-PAGE and activity staining with 4-MUG substrate showed that both A333 and A343 had a differential enyzme band in addition to a common band showing the same electrophoretic mobilities with the standard strains. α-Glucosidase was then eluted from N-PAG bands and seperated by 10% SDS-PAGE, and results showed that Bacillus sp. A333 and A343 isolates had two different protein bands having approximately 44 and 60 kDa molecular weight. A333 α-glucosidase showed optimum activity at 60ºC, pH 6.8 and 1.38 Km value for pNPG substrate while these results were found 65ºC, 7.0 and 0.85, respectively for isolate A343. Specificity for 20 different substrates and Thin layer chromatography of enzymes demonstrated that A333 enzyme had high transglycosylation activity while A343 had wide substrate specificity. In addition, the results showed that Bacillus sp. A333 α-glucosidase was more stable in a wide range of pH and temperature, and both enzymes were found to be resistant to the most of the inhibitors tested. By this study, two new thermostable exo-α-1,4-glucosidase producer Bacillus strains were isolated having biotechnological pottential.
