dc.description.abstractBu araştırma, ayva nektarındaki biyoaktif bileşenler ve antioksidan aktivitenin depolama süresince değişimini belirlemek amacıyla yapılmıştır. Ayva nektarı örnekleri 5°, 20°, 30° ve 40 °C'de 9 ay süreyle depolanmış ve her ay analiz yapılmıştır. Antioksidan aktivitenin belirlenmesinde TEAC ve DPPH yöntemleri kullanılmıştır. Toplam fenolik madde, flavonoid madde ve L-askorbik asit miktarları ise spektrofotometrik yöntemlerle belirlenmiştir. Depolama başlangıcındaki 788.44 mg GAE/L olan toplam fenolik madde miktarı, depolama sonunda 5°, 20°, 30° ve 40 °C sıcaklıklarda sırasıyla 669.2, 605.0, 559.3 ve 450.8 mg GAE/L olarak saptanmıştır. Depolama sonunda flavonoid madde miktarında 5°, 20°, 30° ve 40 °C sıcaklıklarda sırasıyla %12.77, % 24.93, %26.83 ve %34.69 azalma belirlenmiştir. Depolama başlangıcında TEAC yöntemine göre belirlenen antioksidan aktivite düzeyi 266.2 µmol TE/100g iken DPPH yöntemine göre belirlenen antioksidan aktivite düzeyi 123.1 µmol TE/100g'dır. Antioksidan aktivite düzeyinde depolama süresince tüm sıcaklıklarda önemli düzeyde bir azalma gözlenmiştir. Depolama sonunda askorbik asitte 5°, 20°, 30° ve 40 °C sıcaklıklarda sırasıyla %32.08, %43.69, %65.21 ve %88.82'lik bir azalma belirlenmiştir. Askorbik asit degradasyonunun birinci dereceden tepkime kinetiğine uyduğu ve aktivasyon enerjisinin 43.65 kj mol-1 olduğu saptanmıştır. Depolama süresince en çok HMF oluşumu % 369.89 artış ile 40 °C'de depolanan örneklerde bulunmuştur. HMF oluşumunun sıfırıncı dereceden tepkime kinetiğine göre geliştiği ve aktivasyon enerjisinin 88.303 kj mol-1 olduğu belirlenmiştir. Ayva nektarlarının antioksidan aktivitesi üzerine toplam fenolik madde, flavonoid madde ve L-askorbik asidin etkisi de incelenmiştir. TEAC ve DPPH yöntemiyle belirlenen antioksidan aktivite ile L-askorbik asit, toplam fenolik madde ve flavonoid madde miktarları arasında önemli bir korelasyon saptanmıştır. Ayrıca TEAC ve DPPH yöntemleri arasında da önemli bir korelasyon (r =0.855 p<0.01) belirlenmiştir.AbstractThis study was conducted to determine the changes of bioactive components and antioxidant activity of quince nectar during storage. Quince nectar samples have been stored at 5°, 20°, 30° and 40 °C for 9 months and analyses were carried out each month. Antioxidant activity was determined using TEAC and DPPH assays. The content of total phenolic, flavonoid and L-ascorbic acid were determined by spectrophotometric methods. At the beginning of storage, the content of total phenolic was 788.44 mg GAE/L and at the end of storage it was found as 669.2, 605.0, 559.3 and 450.8 mg GAE/L at the storage temperatures of 5°, 20°, 30° and 40 °C, respectively. The loss of flavonoid content at the end of storage was found as 12.77 %, 24.93 %, 26.83 % and 34.69 % at the storage temperatures of 5°, 20°, 30° and 40 °C, respectively. At the beginning of storage, while the content of antioxidant activity by TEAC assay was 266.2 µmol TE/100g, the content of antioxidant activity by DPPH assay was 123.1 µmol TE/100g. A significant decline of antioxidant activity could be observed during storage at all temperatures. Loss of ascorbic acid after storage at 5°, 20°, 30° and 40 °C was 32.08 %, 43.69 %, 65.21 % and 88.82 %, respectively. The loss of ascorbic acid followed a first-order kinetic model and activation energy was determined as 43.65 kj mol-1. The highest HMF accumulation with the increase of 369.89 % was found in the samples stored at 40 °C. HMF accumulation fitted to a zero-order kinetic model and activation energy was found as 88.303 kj mol-1. The effects of total phenolics, flavonoids and L-ascorbic acid on antioxidant activity of quince nectars were also evaluated. A significant correlation was found between L-ascorbic acid, total phenolic, flavonoid and antioxidant activity as determined by TEAC and DPPH assays. A significant correlation between TEAC and DPPH assays was also determined.
