Tunceli sarımsağında (Allium tuncelianum (Kollman, N.Özhatay, D. Matthew, Ş. Şiraneci) in vitro kök ve sürgün ucu kültürü yoluyla çoğaltma
Özet
Bu araştırmada, ülkemize özgü bir tür olan Tunceli sarımsağını, doku kültürü tekniklerinden kök ucu kültürü ve sürgün ucu kültürü yöntemleriyle daha kısa sürede ve yoğun bir şekilde çoğaltabilme olanakları belirlenmeye çalışılmıştır. Kök ucu kültürü çalışmalarında köklerde kallus oluşumu sağlanamamıştır. 0.1 mg/l BA ile 1.0 mg/l NAA bulunan ortamda hafif kallus başlangıcı görülse de daha sonra hiçbir gelişme görülmemiştir. Kök ucu kültürü çalışmamızda beklenen başarıya ulaşılamamıştır. Sürgün ucu kültürü çalışmasında sürgün uçlarının dikiminden yaklaşık 1 ay sonra kardeş oluşumu başlamıştır. 4 alt kültür sonunda kardeş sayısı bakımından IAA bulunan ortamların NAA bulunduran ortamlardan daha başarılı sonuçlar verdiği belirlenmiştir. Büyümeyi düzenleyiciler kardeşlenme oranı açısından karşılaştırıldığında ise en yüksek kardeşlenme oranı %76 ile 0.05 mg/l BA+0.1 mg/l IAA bulunan ortamdan alınmıştır. Bunu %67 ile 0.1 mg/l BA, 0.1 mg/l BA+0.1 mg/l NAA ve %65 ile 0.05 mg/l BA+0.5 mg/l IAA, %56 ile 0.05 mg/l BA bulunan ortamlar izlemiştir. Bu türün çoğalma katsayısının düşük olduğu ve sürgün başına yaklaşık 1 adet kardeş elde edildiği belirlenmiştir. Baş oluşumunda 0.1 mg/l NAA'in IAA'e göre daha etkili olduğu ve alt kültür sayısı artıkça sürgünlerde baş oluşumunun da arttığı ortaya konulmuştur.Abstract In this research, possibilities for rapid propagation in shorter times for Tunceli garlic, which is endemic to our country, were determined by root culture and shoot tip culture methods which are among rapid propagation and tissue culture techniques. Callus formation couldn?t be provided in the roots in root tip culture studies. Although slight callus inception was observed in the environment where there was 0.1 mg/1 BA and 1.0 mg/1 NAA, no advance was observed afterwards. Expected success was not achieved in the root tip culture study. In the shoot tip culture study, shoot formation started approximately 1 month after shoot tip planting. After 4 sub-cultures, it was determined that environments containing IAA gave out better results than environments containing NAA with regard to the number of shoots. When environments are compared with regard to shoot rates, the highest shoot rate was obtained from the environment containing 0.05 mg/l BA+0.1 mg/l IAA with 76%. This was followed by environments containing 0.1 mg/l BA, 0.1 mg/l BA+0.1 mg/l NAA with 67% and 0.05 mg/l BA+0.5 mg/l IAA with 65%, 0.05 mg/l BA with 56%. It was determined that the propagation co-efficient of this species is low and approximately 1 shoot can be obtained per shoot. Of the auxins, NAA was shown to be more efficient in bulb formation in shoots and bulb formation increased along with the increase in the number of sub-cultures.
