Serin alkali proteaz sentezini kontrol eden aspartik asit grubu amino asitlerin proteaz üretimine etkisinin araştırılması

Serin Alkali Proteaz (SAP) üretiminde subC içeren plasmid pHV1431 taşıyan rekombinant Bacillus licheniformis ile SAP sentezini kontrol eden aspartik asit grubu amino asitlerin ?aspartik asit (Asp), asparajin (Asn), izolözin (Ile), lizin (Lys), treonin (Thr), metionin (Met)- SAP üretimine etkisinin belirlenmesi amacıyla farklı ekleme zamanlarında ve CAA=0-15 mM derişim aralığında etkileri incelenmiştir. Bu amino asitlerden Asp, Asn, Ile, Thr, Lys sırasıyla te=0 ekleme anında CAsp=7.5 mM derişiminde 1.35 kat, te=18 ekleme anında CAsn=5.0 mM derişiminde 1.38 kat, te=18 ekleme anında CIle=15 mM derişiminde 1.25 kat, te=18 ekleme anında CThr=7.5mM derişiminde 1.32 kat, te=0 ekleme anında CLys=2.5 mM derişiminde 1.12 kat attırırken, metionin (Met) ise üretimi inhibe etmiştir. Diğer taraftan farklı rekombinant türleri ile de birer set deney yapılmış, rekombinant B.subtilis ile te=0 ekleme anında üç farklı derişimde (2.5, 5.0, 7.5 mM), rekombinant B.amyloliquefaciens ve rekombinant B.sphaericus ile de te=0'da ve her biri CAA=2.5 mM derişimde aspartik asit grubu amino asitlerin etkisi incelenmiş; bunun sonucunda rekombinant B.subtilis'de Asp 1.28 kat, Asn 1.25 kat, Thr 1.30 kat, Lys 1.67 kat, Met 1.41 kat üretimi arttırmış; Ile ise inhibe etmiştir. Rekombinant B.amyloliquefaciens'de ise Asp üretimi 1.35 kat, Asn 1.25 kat arttırmış; Thr, Ile, ve Lys ise inhibe etmiştir. Rekombinant B.sphaericus'da ise Asp üretimi 1.35, Thr 1.28 kat, Lys 1.26, Ile ise 1.22 kat arttırmış; Asn ise üretimi inhibe etmiştir. Programın ikinci aşamasında ise rekombinant B.licheniformis ile optimum derişimleri belli olan aspartik asit grubu amino asitlerin SAP verimine ve seçimliliğine, ürün ve yan ürün dağılımlarına etkisi incelenmiş ve her amino asit için üç farklı derişimde metabolik yolizi analizi yapılmıştır. Metabolik yolizi analizinin sonucunda SAP üretiminde ortama optimum ekleme anında ve optimum derişimde eklenen Asp, Asn, Thr, Ile, Lys'nin SAP üretimini arttırarak üretimde bir darboğaz olmaktan çıktığı belirlenmiş, bu çalışmadan sonra yapılacak çalışmalarda mikroorganizmalarda regülasyon mekanizmaları dikkate alınarak genetik modifikasyon yapılması durumunda üretimin daha fazla arttırabileceği sonucuna ulaşılmıştır.AbstractThe effects of concentration and addition of aspartic asid group amino acids on serine alkaline protease (SAP) production in order to determine the controlling amino acids of SAP synthesis were investigated by using recombinant Bacillus licheniformis carrying subC gene encoding SAP enzyme. SAP activity was increased in each enriched bioprocess medium containing 7.5 mM aspartic acid (Asp), 5.0 mM asparagine (Asn), 15 mM ısoleucine (Ile), 7.5 mM threonine (Thr), 2.5 mM lysine (Lys), respectively, 1.35, 1.38, 1.25, 1.32, 1.12 fold. Methionine was inhibated the production of SAP synthesis. The effects of aspartic acid group amino acids on SAP production with he other recombinant Bacillus species (r-B.subtilis, r-B.sphaericus, r-B.amyloliquefaciens) was investigated. With r-B.subtilis; SAP activity was increased in each enriched bioprocess medium containing Asp, Asn, Thr, Lys, Met respectively 1.28, 1.25, 1.30, 1.67, 1.41 fold but Ile inhibated the production of SAP synthesis. With r-B.sphaericus; SAP activity increased in each enriched bioprocess medium containing Asp, Thr, Lys, Ile respectively, 1.35, 1.28, 1.26, 1.22 but Asn was inhibated the SAP production. With r-B.amyloliquefaciens SAP activity increased in each enriched bioprocess medium containing Asp, Asn respectively 1.35, 1.25 fold but Ile, Lys, Thr inhibated the SAP production. Threafter, the mass-flux balance based stoichiometric model for the bioreaction network that includes 236 reactions and 183 metabolites were developed; and then solved using the extracellular concentration data together with the batchbioreactor medium concentration measurements. Results of the metabolic pathway analysis; aspartic acid group amino acids were bootleneck of the SAP production.
