Türk toplumunda GSTP1 gen polimorfizminin incelenmesi
Özet
Glutatyon S-transferazlardan biri olan GSTP1, ksenobiyotik metabolizmasında ve ilaç rezistansında rol oynamaktadır. GSTP1' in İle105Val ve Ala114Val gibi polimorfizmleri enzim kodlanma fonksiyonunda değişikliklere ve böylelikle kişiler arasında çeşitli kanser türlerinde ve kemoterapiye yanıtta farklılıklar meydana getirmektedir. Türk popülasyonunda bu polimorfizmlere ilişkin çalışmalar az sayıda olup bazıları çelişkilidir. Böylelikle Türk popülasyonunda GSTP1 gen polimorfizmlerinin frekanslarını belirlemeyi amaçladık. GSTP1 polimorfizm frekansları 233 sağlıklı gönüllüden alından kan örneklerinden izole edilen DNA' larda yürütülen polimeraz zincir reaksiyonu ve restriksiyon fragmenti uzunluk polimorfizmi (PCR-RFLP) yöntemleri kullanılarak saptandı. Ekzon 5 polimorfizm frekansları, İle105İle için %54,1; İle105Val için %35,6 ve Val105Val için %10,3 olarak; GSTP1 ekzon 6 polimorfizm frekansları ise, Ala114Ala için %84,1; Ala114Val için %14,2 ve Val114Val için %1,7 olarak bulunmuştur. Bu sonuçlar Türk popülasyonunda daha önce yapılmış bazı çalışmalarla uyum, Avrupa beyaz ırk toplumlarında yapılmış çalışmalardan bazılarıyla benzerlik göstermektedir. Bununla beraber bu çalışmada elde edilen ekzon 5 polimorfizmi frekansı sonuçları, Hindistan Kore ve Tayvan gibi doğu Asya toplumlarından farklı(azdır).Abstract GSTP1, one of Glutathione S-transferases, is involved in xenobiotic metabolism and drug resistance. Polymorphism of GSTP1 Ile105 Val and Ala114 Val may account for the alteration function of the coding enzyme and thus cause interindividual differences in certain types of cancers and response to chemotherapy. Studies regarding these polymorphisms in Turkish population is rare and some of them are contradictory. Thus, we aimed to clarify the frequencies of GSTP1 gene polymorphisms in a Turkish population. The frequencies of GSTP1 polymorphisms were determined by using the polymerase chain-reaction-restriction fragment length polymorphism (PCR-RFLP) in the DNAs isolated from the blood samples of 233 healthy volunteers. The frequencies of GSTP1 exon 5 polymorphism were Ile105Ile: 54,1%, Ile105Val: 34,3%, Val105Val: 11,6% and GSTP1 exon6 polymorphism were Ala114Ala: %84,1, Ala114Val: %14,2 and Val114Val: %1,7. These results are consistent with some of the previous studies performed in Turkish population and similar to those of European Caucasian populations. However the results of this study regarding the frequency of exon 5 polymorphism is different (lower) from east asia countries such as India, Korea and Taiwan .
