İndol türevi bazı yeni bileşiklerin sentezleri ve anti-kanserojen etkilerinin araştırılması
Özet
Kanser, hücrelerin kontrolsüz çoğalması ve bunun sonucu olabilen metastaz ile kendini gösteren, çağımızın en önemli ve dramatik mediko sosyal sorunlarından biridir. Tirozin kinazların kanser oluşumu üzerindeki rolünün anlaşılması ile bu enzimlerin kanser kemoterapisinde evrensel hedef olarak kabul edilmiştir. Buna ek olarak kanser hücrelerinde reseptörsüz Src tirozin kinaz enzimlerinin kontrolsüz aktivasyonuyla görülen tümör büyümesi ve metastazdaki hızlanma, Src tirozin kinazların da kanser için ilaç tasarımı araştırmalarında evrensel ve önemli bir hedef olmasına yol açmıştır. Ancak günümüzde tedavi amacıyla bir src tirozin kinaz inhibitörü kemoterapötik olarak piyasaya sürülmemiştir. Bu tez çalışması kapsamında amaç Src tirozin kinaz inhibitörü olabileceğini düşündüğümüz indol halkası taşıyan, N-benzil indol-3-imin, N-benzil 5-bromo indol-3-imin, N-benzil 5-fenil indol-3-imin, N-benzil 5-(p-florofenil) indol-3-imin ve N-benzil indol-3-amin, N-benzil 5-bromo indol-3-amin, N-benzil 5-fenil indol-3-amin, N-benzil 5-(p-florofenil) indol-3-amin yapısında olan kırk adet bileşik tasarımı ve sentezlenmesi ve src tirozin kinaz hedefine karşın aktivitelerinin incelenmesidir. Bu amaçla hedef bileşiklerin sentezi için, başlangıç maddesi olarak indol ve 5-bromo indol yapısındaki bileşikler seçilmiştir. Kimyasal yapıları ve saflıkları IR, 1H-NMR, kütle ve elementel analiz sonuçları kanıtlanmıştır. Bu tez çalışmasında elde edilen bileşiklerin saflıkları spektral yöntemlerle kontrol edildikten sonra erime noktaları saptanmıştır. Sentezlenen bileşiklerin Src tirozin kinaz inhibitör aktivitesi ise ELISA yöntemiyle evrensel tirozin kinaz enzim kiti kullanılarak gerçekleştirilmiştir. Sentezlenen bileşiklerden imin yapısındakilerde Src inhibitör aktivitesi gözlenmemiştir. Diğer taraftan amin yapısında olan türevlerden dördünde aktivite tespit edilmiştir. Aktif olan 2c-3, 2d-1, 2d-2 ve 2d-3 bileşikler için, sırasıyla, 4,69±1,23 µM, 74.79±1,43 µM, 75.06±1,24 µM ve 84.23±1,19 µM IC50 değerleri bulunmuştur. Bu sonuçlar ışığında, aktif bileşiklerden 2c-3 bileşiği ise en düşük IC50 değeriyle ilaç geliştirme çalışmaları açısından Src tirozin kinazı hedefleyen umut verici bir bileşik olarak tespit edilmiştir. abtractCancer is the most important and dramatic medico-social problem nowadays, with cell proliferation, uncontrolled cell growth and hence metastasis. After understanding the role of tyrosine kinases on cancer development and progress, inhibitors of tyrosine kinase enzymes became universal targets for cancer chemotherapy. Moreover, increased rate of tumor growth and metastasis with uncontrolled activation of non-receptor Src tyrosine kinases, resulted that Src tyrosine kinases are the important and universal targets for anticancer drug design research. However, there is stil no Src tyrosine kinase inhibitor reached to the market as anticancer chemotherapeutics. In this thesis, it is aimed to design and synthesize forty indole ring bearing compounds as derivatives of N-benzyl indole-3-imine, N-benzyl 5-bromo indole-3-imine, N-benzyl 5-phenyl indole-3-imine, N-benzyl 5-(p-fluorophenyl) indole-3-imine ve N-benzyl indole-3-amine, N-benzyl 5-bromo indole-3-amine, N-benzyl 5-phenyl indole-3-amine, N-benzyl 5-(p-fluorophenyl) indole-3-amine, as possible Src tyrosine kinase inhibitors. It is aimed to analyze the activity of synthesized compounds, against non-receptor Src tyrosine kinase target. Here, for the synthesis of targeted compounds, indole and 5-bromo indole were chosen as the starting compounds. Chemical structures and the purity of the resulted compounds were elucidated by IR, 1H-NMR, MASS spectroscopy, and the elemental analysis. In this study, the melting points of the synthesized compounds were determined after their purity were analyzed by spectral analyzes. The Src tyrosine kinase inhibitor activity of the synthesized compounds was determined via ELISA method by using the universal tyrosine kinase assay kit. Among the compounds synthesized, the imine derivatives were found with no inhibitor activity against Src tyrosine kinase target. On the other hand, four of the amine derivatives found as active Src tyrosine kinase inhibitors. The IC50 values for active compounds, 2c-3, 2d-1, 2d-2 and 2d-3, were determined as 4,69±1,23 µM, 74.79±1,43 µM, 75.06±1,24 µM ve 84.23±1,19 µM, respectively. In the light of these results, the compound 2c-3 with lowest IC50 was found the promising compound targeting Src tyrosine kinase for drug discovery efforts.
