Amlodipin ve rosuvastatin'in karışımlarında LC-MS/MS ile aynı anda miktar tayinleri

Amlodipin ve Rosuvastatin'in Karışımlarında LC-MS/MS ile Aynı Anda Miktar Tayinleri Yapılan literatür araştırmaları sonucunda AML ve ROS etken maddesinin birarada tayinine ilişkin herhangi bir LC-MS/MS çalışmasına rastlanmamıştır. Bu nedenle bu çalışmada bu iki maddenin eşzamanlı tayinine ilişkin basit, hızlı, kesinliği yüksek, tayin edilebilir sınırı düşük bir LC-MS/MS yöntemi geliştirilmesi ve geliştirilen bu yöntemin tabletlere ve idrar numunelerine uygulanması amaçlanmıştır. Tez çalışmamız kapsamında metod optimizasyonunu yapabilmemiz için incelenen faktörler çözücü sistemi içindeki organik çözeltinin oranı (B1), tampon çözeltideki amanyum format konsantrasyonu ve pH' dır. İncelenen bu faktörlerin analize etkisinin ve bu faktörlerin metodu optimize edebilmek için bir deneysel tasarım tekniği oluşturulması amaçlanmıştır. Bu amaç doğrultusunda merkezi kompozit tasarım tekniğinin seçilmesi uygun görülmüştür. Geliştirilen yöntem kolon olarak ACQUITY HSS-C18, 2.7 μm, 2,1x150 mm, mobil faz olarak (pH: 5.00, 15mM tampon çözeltisi) (%1'lik formik asitli asetonitril) ( %8: %92), 10,00 μL' lik enjeksiyon hacminde 0,4 mL/dk akış hızı ile çalışılmıştır. Kütle spektrometresi analizleri Waters TQD LC-MS/MS sistemi kullanılarak gerçekleştirilmiştir. Alıkonma zamanları ise AML için 0,85 ve ROS için 0,88 dakika olarak bulunmuştur. Kullanılan ekstraksiyon yönteminin (KFE) ortalama yüzde geri kazanımı AML ve ROS için sırasıyla %99,68, %100,10 olarak bulunmuştur. Yöntemde ki yakalama sınırı (YS) AML ve ROS için sırasıyla için 0,61 ve 0,91 ng/mL; tayin alt sınırı (TAS) AML ve ROS için sırasıyla 1,97 ve 2,50 ng/mL olup, yöntemin AML için 2,50 - 100,00 ng/mL ve ROS için 2,50 - 100,00 ng/mL değerleri arasında doğrusallık gösterdiği bulunmuştur. Geliştirilen yöntem farmasötik preparatlara ve etken maddelerin katıldığı idrar örneklerine başarıyla uygulanmıştır. AbstractThere is no reports for simultaneous determination of AML and ROS with LC-MS/MS in the literature. Therefore we aimed to develop a simple, fast, precise, sensitive LC-MS/MS method for simultaneous determination of these two substances. It was aimed to apply the method to tablets and urine samples. In this study, we focused on organic solvent ratio (B1) in solvent system, ammonium formate concentration and pH in buffer solution parameters for method optimization. We aimed to investigate effects of these parameters on analysis and developed the experimental technical for optimization of this method by using these parameters. We selected central composite design. In the developed method, ACQUITY HSS-C18, 2.7 μm, 2,1 x 150 mm column was used as the stationary phase and (pH: 5.00, 15mM buffer solution) (1% formic acid containing acetonitrile) (8%:92%) were used as the mobile phase. Injection volume was 10 μL while flow rate was 0.4 mL/dk. Mass spectrums were taken with Waters TQD LC-MS/MS system. Retention times for AML and ROS were 0.85 and 0.88 minutes respectively. The recovery of the extraction method (SPE) was for amlodipine 99,68% and for rosuvastatin 100,10%. In relation with the method; limit of detection (LOD) for AML and ROS are 0,61 and 0,91 ng/mL, respectively, limit of quantitation (LOQ) for AML and ROS are 1,97 and 2,50 ng/mL, respectively in the standarts. The method has been found to exhibit linearity for amlodipine between the values 2,50-100,00 ng/mL and for rosuvastatin between the values 2,50-100,00 ng/mL The developed method has been successfully applied to pharmaceutical preparations and urine samples in which active ingredients are involved.
