Canefe, KandemirÖz, Umut Can2022-06-222022-06-222011http://hdl.handle.net/20.500.12575/82213The main purpose of this study is to investigate and regulate the optimization parameters of the diclofenac sodium loaded microparticulate systems such as microspheres which used in formulations of the aqueous dispersions and produced by w/o/w and s/o/w emulsion - solvent evaporation techniques.Two different polymers were used at in microsphere formulations which were produced with chosen production method. The production yield, encapsulation efficiency, particle size and size distribution, FTIR analyses, DSC analyses, optical microscope analyses either with fluorescence dedector or not, zta potential measurements, micro-CT investigations, residual DCM dedection, studies on stability tests and in-vitro drug release experiments were conducted on the microsphere formulations which were obtained with Eudragit RS 100 and Poly-?-caprolactone polymers.Viscosity and pH alteration analyses, sedimentation analysis, particle size and appearance alteration investigation on suspended microspheres, alteration of encapsulation efficiency after suspension were observed on aqueous dispesion formulation which were produced with obtained microspheres.After evaluation the microsphere formulations, microsphere formulation F2 was chosen on the grounds of investigated parameters results. The second step of the study was producing an appropiate aqueous dispersion formulation to deliver diclofenac sodium loaded micospheres by perorally. In order to achieve this step reconstitute suspension form was produced with the excipients like xanthan gum, D-sorbitol, citric acid, sodium benzoate, sodium lauryl sulphate, which was chosen as aqueous dispersion formulation.In conclusion, stable and optimized aqueous dispersion that carries diclofenac sodium loaded microparticles was produced during storage and usage.
