In our country, drugs and pesticides which cause often poisoning connected with quantative to be developed was nec- cessary. In poisoning, as they were the most common drugs and pesticides have chosen. These were which in structure of acid : acetaminofen, acetyl salicylic acid, salicyl acid, phenobarbital, pentobarbital, which in structure of basic : amitriptyline, nor - triptyline diphenhydramine, imipramine, clomipramine, chlor - promazin, codein, nicethamide, which in structure of neutral : phenylamine maleate, methocarbamol, meprobamate, and so me pesticides (diazinone, malathion, ethyl parathion, prope- tamphos, sumithion, trichlorfos. These drugs and pesticides to isolation from postmortem tissue (fresh sheep liver) enzymatic digestion, acidic hydrolysis, precipiçaüon, Stas - Otto of methods as compared hava been studied. Papain and trypsin as proteolytic enzyme have been used to increase the extraction procedure yield in enzymatic di gestion. The methods of liquid - liquid extraction and Amberli- te XAD-2 Bacth have been used to be taken away the conside rable analytical background which can be interfered. The isolation of drugs and pesticides could be identified by means of TLC and UV spectrofotometry. In this study, as the first screening test thin layer chro-121 matography is used because of its simplicity, rapidity, and its cheapness. The optimum contidions of confirmatory and quantative determinations of these drugs and pesticides were determina ted on GLC. Reasonable seperations have been achieved in two different colum systems ( % 3 SE-30 and % 3 OV-17) using FID detector. The oven temparature 180° - 220 °C and the flow rate (30-50 ml/min.) of nitrogen have been a good seperation by changing. The avarage yield of extraction and isolation have been obtained and compared by several of methods in the GLC. The recoveries of the drugs and pesticides for enzymatic digestion papain : % 73.9 ± 1.7; trypsin : % 66.3 ± 2.2 precipitation; % 46.5 ± 1.8 acid hydrolysis : % 38.5 ± 2.9 Stas - Otto : % 27.6 ± 2.4 have been obtained.
