Abstract
The WUSCHEL-Related Homeobox (WOX) gene family, a class of homeobox transcription factors, plays a role in plant-specific embryonic modeling and important developmental processes such as embryonic development, stem cell care, organ and seed formation and regeneration of isolated tissues and organs. In this thesis, the identification and characterization of the members of the WOX gene family, which are transcription factors, was carried out by using a number of bioinformatics databases and programs in order to illuminate the contribution to evolutionary processes and to benefit the field of plant biotechnology. A total of 18 WOX (Phvul-WOX) genes belonging to the bean genome have been identified. Sequences alignment, determination of conserved motifs, homology modeling of proteins, gene duplications, homologous and non-homologous change rates were determined. Besides these; exon-intron analysis, phylogenetic analysis, analysis of promoter regions, detection of extracellular localization rates, miRNA analysis, synteny analysis, tissue-specific mRNA and in silico expression analysis of genes associated with salt stress were performed. The phylogenetic tree created is divided into three classes as WUS, intermediate and old. The reliability of the tree was verified by motif identification and exon-intron. In silico expression analysis of the identified Phvul-WOX genes in the leaf tissue of the plant and after salt stress treatment of the target genes, mRNA expression levels were determined by qPCR reactions, normalization and statistical analysis of the data were performed. Thus, this study is the first in terms of identification of WOX genes in P. vulgaris species and determination of their relationship with salt stress. It is a resource for future studies.
