Bademin (Prunus amygdalus Batsch) in vitro vegetatif çoğaltımında besin ortam içeriğinin sürgün verimine etkileri
Abstract
In this research the possibilities of in vitro vegetative propagation of almond cv. Texas and Nonpareil by shoot tip culture were investigated. For this purpose four experiments were established with different sucrose, agar and pH levels and IBA+BAP combinations. In these experiments proliferation, shoot production and development were tested in all three stages culture. In initiation stage proliferation was only achieved in sucrose experiment with 5-6 % sucrose. In this stage the best explant development was obtained from 0.5 % agar, 5.5 pH and 0.0-0.1 mg/1 IBA+0.0 mg/1 BAP combinations. In transplantation stage, 5-6 % sucrose, 5.5 pH levels and 0.1 mg/1 IBA and 1.0 mg/1 BAP concentration gave the best results with respect to shoot production and development. The medium with low agar concentrations was favourable for shoot formation and development however, since these levels caused vitrification 0.7 % agar dose was determined to be most appropriate.vx In multiplication stage, we obtained a good rate of shoot multiplication from 3-4 % sucrose, 0,7 % agar, 5.5 pH and 0.1 mg/1 IBA+1.0 mg/1 BAP combination. In all IBA doses, high BAP (2.0-3.0 mg/1) concentrations induced early proliferation, but they gave rise to vitrification, fasciation, leaf deformation and callus formation as well. In addition, cytokinin (BAP) is definitely essential for shoot proliferation in transplantation and multiplication stages.KEY WORDS : In vitro, shoot tip culture, Prunus amygdalus Batsch, almond, micropropagation.
