Fotoakustik mikroskop sistemi için görüntü oluşturma
Abstract
Fotoakustik mikroskop (Photoacoustic Microscopy, PAM), fotoakustik etki temeline dayanan ve son yıllarda sıkça çalışılan hibrid bir görüntüleme tekniğidir. Optik kontrastı akustik olarak algılayan yapısı sayesinde sistem, optik difüzyon sınırının ötesinde derin bir görüntülemeyi yüksek çözünürlükle sunabilmektedir. PAM sistemi ile görüntülemede kaydedilen sinyaller, sistem bileşenleri ve çevresel etkilerle gürültüye maruz kalmaktadır. Yapılan çalışmada PAM sisteminde kullanılmak üzere bir görüntü oluşturma algoritması önerilmiş, algoritmanın filtreleme adımında, üç farklı grupta filtreleme çalışmaları yürütülmüştür. İlk grupta akustik dalga denkleminin çözümüyle oluşturulan akustik sinyal üzerine belirli oranlarda sentetik gürültüler eklenmiştir. Gürültülü sinyallere FIR alçak ve bant geçirgen filtreler yanısıra bior3.5, bior3.7 ve sym7 dalgacıkları ile ayrık dalgacık dönüşümü tabanlı filtreleme uygulanmış ve filtreleme performansının değerlendirilmesi için sinyaller üzerinde kalite metrikleri hesaplanmıştır. İlk grupta elde edilen veriler doğrultusunda, ikinci grupta test cismi sinyallerinin filtrelenmesi ve üçüncü grupta kan hücresi sinyallerinin filtrelenmesi için alçak ve bant geçirgen filtreler ile birlikte, ayrık dalgacık dönüşümü tabanlı filtreleme için bior3.7 ve sym7 ana dalgacıkları seçilmiştir. Elde edilen filtreli sinyallerden görüntüler oluşturulmuş ve oluşturulan görüntüler üzerinde filtre performanslarının değerlendirilebilmesi için metrikler hesaplanmıştır. Sonuçlar incelendiğinde, ayrık dalgacık dönüşüm tabanlı filtrelemede sym7 dalgacığı minimaxi yumuşak eşiklemenin çok yüksek gürültü seviyelerinde, sym7 sqwotlog yumuşak eşiklemenin yüksek ve orta gürültü seviyelerinde, alçak geçirgen filtrenin düşük gürültü seviyelerinde ve bant geçirgen filtrenin de çok düşük gürültü seviyelerinde başarılı olduğu görülmüştür.
Photoacoustic microscopy (PAM) is a hybrid imaging technique based on the photoacoustic effect and frequently studied in recent years. Through the system structure that senses the optical contrast acoustically, it can present a deep image at a high resolution beyond the optical diffusing limit. Signals recorded with the PAM system are exposed to noise by system components and environmental influences. In this study, an image reconstruction algorithm is proposed for use in the PAM system. In the filtering step of the algorithm, three different group of filtering were carried out. In the first group, a synthetic noise is added at a certain ratio to the acoustic signal generated by the solution of the acoustic wave equation. Noisy signals were filtered by FIR low and band pass filters as well as discrete wavelet transform based filtering using bior3.5, bior3.7 and sym7 mother wavelets and quality metrics were calculated on the signals for evaluation of filtering performance. In the second group for filtering the test material signals and in the third group for filtering the blood cell signals bior3.7 and sym7 mother wavelets were selected for discrete wavelet transform based filtering, along with low-pass and band-pass filters. Then, images were generated using the filtered signals and metrics were calculated to evaluate the filter performances on the generated images. As a result, it was found that for very high noise levels the sym7 minimaxi soft thresholding, for high and medium noise levels the sym7 sqwotlog soft thresholding, for low noise levels the low-pass filter and for very low noise levels the band-pass filter gave the best results.
