Organik özütlerle priming uygulamalarının soğan (allium cepa) ve havuç (daucus carrota) tohumlarının abiyotik stres koşulları altında çimlenme ve fide kalitesine etkisi

Kuraklık, tuzluluk ve yüksek sıcaklık, bitki gelişimi ve verimliliğini engelleyen önemli abiyotik streslerdir. Tohum çimlenmesi ve fide gelişimi, abiyotik strese karşı bitkinin en hassas olduğu aşamalardır. Bu nedenle, ekimden önce tohumların kalitesini artırmak, strese tolerans sağlamak için önemlidir. Bu çalışmanın amacı organik özütlerle priming uygulamalarının havuç (Daucus carota) ve soğan (Allium cepa) tohumlarının abiyotik stres koşulları altında çimlenme ve fide kalitesine etkisini araştırmaktır. Uygulamalarda deniz yosunu (%5 DY), butenolid (10-7 But) ve vermikompostun (%5 VC) tekli, çiftli ve üçlü kombinasyonları her iki türde de yüksek ve düşük kalitedeki ikişer tohum partisinde kullanılmıştır. Kuraklık stresi, farklı konsantrasyonlarda % 0, 10, 15 ve 18 PEG-6000, tuzluluk stresi ise, farklı molaritelerde 0, 50, 100, 150 mM NaCl kullanılarak simüle edilmiştir. Sıcaklık stresi ise tohumların 20, 30, 35 ve 40°C’de çimlendirilmeleriyle test edilmiştir. KMP uygulaması tohum: vermikülit:su 2:1:3 oranında soğan için 3 gün havuç için de 5 gün 15ºC’de karanlıkta yapılmıştır. Bütün katı matris priming işlemi, her iki türün tohumlarının ve fidelerinin, stres koşularında (%10, 15 PEG ve 50, 100 mM NaCl) performansını arttırmıştır. Yüksek düzeydeki stres koşullarında, % 18 PEG, 150 mM NaCl ve 40°C sıcaklık, olumlu etkide bulunmamıştır.
Abiotic stresses such as drought, salinity and temperature extremes are serious problems that hinder crop production and productivity. Seed germination and seedling development are the most sensitive stages to abiotic stress. Thus, enhancing the quality of a seed ahead of sowing is vital to minimize these problems. The present study was aimed to investigate the effect of priming with organic extracts on the quality of seeds and seedlings of carrot and onion under abiotic stress conditions. Seaweed (SW), Butenolide (But) and vermicompost (VC) in single, double and triple combination were used. Drought stress was simulated using PEG-6000 at different concentrations (0, 10, 15 and 18%PEG), while salinity stress was simulated using NaCl at different molarity (0, 50, 100, 150mM), which have same osmotic potential with the corresponding PEG concentrations. At the same osmotic potential, the inhibitory effect of PEG was greater than NaCl. All matrix priming treatment improved performance of seeds and seedlings of both crops at mild but not at very high stress levels. High concentrations of PEG (18%PEG), NaCl (150mM), and very high temperature (40 °C) brought about detrimental effects on seed germination. However, at mild stress, solid matrix priming with seaweed alone and in combination with butenolide significantly (P<0.05) improved the performance of carrot. Likewise, vermicompost alone and its combination with seaweed significantly (P<0.05) improved the performance of onion seeds and seedlings. In low quality seeds of carrot at 10% PEG, maximum
