Patatesten bütanol üretimi üzerine bir çalışma
Abstract
Bu çalışmada, bazı Clostridium türleri ile patatesten aseton, bütanol, etanol üretimi araştırılmıştır. Çalışmalarda C. beijerinckii DSM 6422, C.acetobutylicum DSM 6228, DSM 1731, DSM 792 suşları ve patatesten izole edilerek 16S rRNA dizisine göre Clostridium roseum olduğu anlaşılan suşlar kullanılmıştır. Karbon kaynağı olarak farklı besiyeri oranlarında (50, 100, 200, 300 ve 400 g patates/L), farklı pH değerlerinde (pH 5, 6, 7, 8, 9 ve 10 ), farklı inkübasyon sıcaklıklarında (30, 33 35, 37 ve 39 °C ) ve farklı katkılar eklenerek hazırlanan besiyerinde beş farklı Clostridium suşunun bütanol ve toplam solvent üretim miktarları belirlenmiştir. Bu çalışmalardan elde edilen sonuclarda C. acetobutylicum DSM 6228 suşu ile 400g patatesli ortamda ve 37°C sıcaklıkta 15.02 g/L bütanol ve 22.65 g/L toplam solvent ürettiği saptanmıştır. Aynı besiyerine 1 g sistein-HCl ve 2.5 g glikoz ilavesi ile C. acetobutylicum DSM 792 suşunun bütanol üretiminde % 74.80’ lik artış sağlanarak 8.07 g/L bütanol elde edilmiştir. Patates besiyerine ilave edilen (NH4)2SO4, sistein-HCl, K2HPO4 ve maya özütü toplam solvent oluşumunu artırırken, CaCO3 ve FeSO4 ilavesinin toplam solvent oluşumunu azalttığı belirlenmiştir. Abstract In this study, butanol (acetone butanol ethanol or ABE) was produced from potato in batch cultures using five different Clostridium sp. strains. High butanol production and growth was determined at 400 g.l-1 potato concentration and 37°C temperature for Clostridum sp. strains. The optimal pH for acetone-butanol-ethanol fermentation were found 7 for Clostridum acetobutylicum DSM 1731 and Clostridium roseum, 9 for Clostridium acetobutylicum DSM 792, DSM 6228 and Clostridium beijerinckii DSM 6422 strains. In a conducted experiment, where potato ( 75.45 g.l-1 total sugar obtained from hydrolysis of 400 g.l-1 potato) was used, C.roseum, C.beijerinckii DSM 6422, C.acetobutylicum DSM 6228, C.acetobutylicum DSM 1731 and C.acetobutylicum DSM 792 produced 6.18 g.l-1 butanol with a productivity 0.14 g/g, 6.32 g.l-1 butanol with a productivity 0.17 g/g, produced 10.27 g.l-1 butanol with a productivity 0.19 g/g, 7.84 g.l-1 butanol with a productivity 0.13 g/g and 8.27 g.l-1 butanol with a productivity 0.17 g/g, respectively. The effect of the component concentrations of a synthetic medium on ABE fermentation by Clostridium strains was also investigated. As a result of this study, when 1g.l-1 cysteine- HCl and 2.5 g.l-1 glucose were added into fermentation medium, growth and ABE production of Clostridium sp. strains increased significantly. Butanol production of Clostridium acetobutylicum DSM 792 increased by 74.80% ( from 4.62 g.l-1 to 8.07 g.l-1) as compared to plain medium. The presence of CaCO3 and FeSO4 in fermentation medium total solvent production was inhibited while the presence of (NH4)2SO4, cysteine-HCl, K2HPO4 and yeast extract induced the total solvent production. In conclusion, this study demonstrates that C.acetobutylicum DSM 6228 strain has excellent capacity to convert biomass derived sugars to solvents and can produce over 15.07 g.l-1 (in one case 22.65 g.l-1 ) ABE from potato.
